This study evaluated whether the adapted FCU maintained positive effects on parenting (positive behavior support, limit setting, parental warmth) and child behavioral health (self-regulation, conduct problems, emotional problems). Pediatric (6-12 years) primary care patients with a BMI ≥ 85th%ile (n = 240) were recruited from primary care clinics in Phoenix. Children were 75% Latino, 49% female, and 73% Medicaid recipients. This type 2 effectiveness-implementation hybrid trial compared families randomized to FCU4Health (n = 141) or usual care (n = 99). FCU4Health was delivered over a period of 6 months. This study focuses on a priori secondary outcomes included parenting and child behavioral health targets of the original FCU, assessed at baseline and 3, 6, and 12 months. Significant improvements were found for the FCU4Health condition, compared to usual care, in parenting from baseline to the 3-month assessment [β = .17 (.01; .32)]. Parenting predicted improvements in child self-regulation at 6-months [β = .17 (.03; .30)], which in turn predicted reductions in conduct problems [β = - .38 (- .51; - .23)] and emotional problems [β = - .24 (- .38; - .09)] at 12 months. Ethnicity and language of delivery (English or Spanish) did not moderate these effects. The FCU4Health can improve parenting and child behavioral health outcomes when delivered in primary care.Trial Registration Trial registration number NCT03013309 ClinicalTrials.gov.
Congenital adrenal hyperplasia (CAH) is an autosomal recessive disorder of adrenal steroidogenesis with a broad spectrum of clinical presentations, ranging from the severe classical salt-wasting (SW) and simple-virilizing (SV) form, to the mild nonclassical form. A large variety of CYP21A2 genotypes in correlation with phenotype have been described.

DNA samples from a 14-day-old male newborn with clinical and laboratory signs of SW CAH and family members were subjected for molecular analysis of the nine most common point CYP21A2 mutations by ACRS/PCR method. Direct DNA sequencing of the whole CYP21A2 gene was performed to detect the second mutant allele in the patient. The in silico predicting analysis and the crystal structure analysis of the mutated CYP21A2 protein have been performed.

Molecular analysis confirmed that the patient was compound heterozygote carrying p.Q318X mutation inherited from the mother and a novel c.1271_1279delGTGCCCGCG (p.G424_R426del) variant in exon 10 inherited from the father. The in silico predicting software tools classified the novel mutation as pathogenic. Crystal structure analysis showed that the three residues affected by the novel in-frame deletion form several hydrogen bonds that could lead to impaired stability and function of the CYP21A2 protein. These findings were concordant with the patient's phenotype. The need of several molecular methods to elucidate the genotype in this patient has also been discussed.

Endometrial laminin subunit beta-3 (LAMB3) is a candidate gene whose expression distinguishes the endometrial window of receptivity (WOR) in human. This study aims to examine endometrial LAMB3 levels in patients with repeated implantation failure (RIF), in order to assess the ability of LAMB3 to predict pregnancy outcome.

Endometrial biopsies were taken during the WOR from 21 healthy volunteers in natural menstrual cycles and from 50 RIF patients in mock cycles prior to frozen embryo transfer (FET) cycles. Immunohistochemistry (IHC) staining of LAMB3 was performed, and the H-score was correlated with the pregnancy outcome in subsequent FETs.

In healthy volunteers, endometrial LAMB3 was demonstrated to be highly expressed during the WOR with the staining exclusively in the cytoplasm of the epithelial cells. In a discovery set of RIF patients, the LAMB3 expression level was found to be significantly higher in those who conceived compared to those who did not in subsequent FETs. A receiving operator characteristic (ROC) analysis revealed an area under the curve (AUC) of 0.7818 (95% confidence interval 59.92-96.44%) with an H-score cutoff of 4.129 to differentiate cases with positive or negative pregnancy outcomes. This cutoff achieved an accuracy of 75% in pregnancy prediction in a following validation set of RIF patients, in which the pregnancy rate in subsequent FETs was three-fold higher when the mock cycle LAMB3 H-score was ≥ 4.129 compared to < 4.129.

Microsatellite markers are mainly used to provide updated information on changes in the population structure of geese breeds. The eight goose populations reported 24% private alleles specific for each population. Expected heterozygosity (He) ranged from 0.46 to 0.70. Three breeds were reported highly polymorphic. Inbreeding coefficient (Fis) revealed that three breeds were in a minimum level of extinction danger, while one breed was in a potential endangered situation. Phylogenetic tree, principal component analysis (PCA), and self-organizing map (SOM) were constructed using MATLAB to study the population distribution and relationship among these breeds. Four SNPs were detected, two SNPs at GH gene exon (C123T and C158T), and two SNPs at Pit-1 gene exons (G161A and T282G). Four SNP loci were reported to have a significant effect on geese body weight. They were CT genotype for C123T locus, TT genotype for C158T locus, GG genotype for G161A locus, and GG genotype for T282G locus.In this study, we investigate the release of melanoidin-like product (MLP) from hybrid silica xerogels to control the quantity of MLP in the medium for lead phytoextraction. In the preparation of the hybrid organic-inorganic xerogels with MLP, tetraethoxysilane (TEOS), methyltriethoxysilane (MTES), propyltriethoxysilane (PTES), and 3-aminopropyltriethoxysilane (APTES) were used as precursors. The experimental results suggest that the release of MLP can be easily controlled by partially substituting TEOS with the organosilanes. The addition of the organosilanes lowered the release rate of MLP in the following order of xerogels TEOS, MTES/TEOS, PTES/TEOS, and APTES/TEOS. Furthermore, a novel phytoextraction of lead through the Indian mustard was conducted using the MLP-doped TEOS xerogel. Results show that the addition of TEOS xerogel did not have any influence on the growth of the mustard, whereas the lead uptake significantly increased in a nutrition medium with 1-mM Pb(NO3)2.
