Numerous studies have demonstrated that thioredoxin-interacting protein (TXNIP) expression of peripheral blood leucocytes is increased in coronary artery disease (CAD). However, the molecular mechanism of this phenomenon remained unclear. DNA methylation plays important roles in the regulation of gene expression. Therefore, we speculated there might be a close association between the expression of TXNIP and methylation. In this study, we found that compared with controls, DNA methylation at cg19693031 was decreased in CAD, while mRNA expressions of TXNIP and inflammatory factors, NLRP3, IL-1β, IL-18, were increased. Methylation at cg19693031 was negatively associated with TXNIP expression in the cohort, THP-1 and macrophages/foam cells. Furthermore, Transwell assay and co-cultured adhesion assay were performed to investigate functions of TXNIP on the migration of THP-1 or the adhesion of THP-1 on the surface of endothelial cells, respectively. Notably, overexpressed TXNIP promoted the migration and adhesion of THP-1 cells and expressions of NLRP3, IL-18 and IL-1β. Oppositely, knock-down TXNIP inhibited the migration and adhesion of THP-1 and expressions of NLRP3, IL-18. In conclusion, increased TXNIP expression, related to cg19693031 demethylation orientates monocytes towards an inflammatory status through the NLRP3 inflammasome pathway involved in the development of CAD. © 2020 The Authors. The aim of this study was to conduct a mixed-methods systematic review of psychosis among post-secondary students, including student characteristics, overall prevalence, risk factors, interventions, and their reported experiences. A librarian created a systematic search of nine databases, and quality was assessed using JBI Critical Appraisal Tools. The average age of participants was 21 years, and most identified as female and Caucasian. The prevalence of psychosis was often measured in a way that limited comparison across studies. Risk factors associated with symptoms of psychosis included substance use, depression, and younger age. Other risk factors trending towards a relationship included racial discrimination and anxiety. Protective factors included self-esteem and self-concept clarity. Five interventions were studied, with mixed results, and only three studies reported on the experience of students. While five promising interventions are identified, inadequate testing and replication limit confidence in their effectiveness and there is a notable deficit in qualitative evidence exploring the experiences of these students. Integrating knowledge on risk factors, interventions, and experiences of students with psychosis will help tailor and facilitate their health care while protecting their right to education. © 2020 Australian College of Mental Health Nurses Inc.Consumer-directed, personal recovery-oriented mental health services are now the focus of mental health policies around the globe. However, there has been minimal debate about how the approach applies to young people. This study sought to address this gap through a Delphi approach, using three rounds of surveys, to gain a convergence of opinion from Australian Child and Adolescent Mental Health Service (CAMHS) practitioners regarding the relevance of personal recovery for children and adolescents. The findings suggest that concepts of personal recovery are appropriate for a young population. However, parents and carers are integral to the mental health recovery journeys of young people, either directly as guardian/decision-maker and/or indirectly through the impact of caring for a young person with mental health problems. Further exploration of the relevance and application of personal recovery in the context of young people is warranted, particularly from the perspectives of adolescents, younger children and carers of young people with mental health problems. © 2020 Australian College of Mental Health Nurses Inc.BACKGROUND White blood cell (WBC) counts are used to monitor bone marrow function and to screen for infections. The HemoCue WBC DIFF Point-Of-Care (POC) instrument classifies WBCs through cell image recognition. To evaluate its suitability for monitoring cancer patients, we examined its performance in samples from patient with leukopenia and in samples containing nRBC. METHODS Sysmex samples with WBCs 0.05-3.40 × 109 /L were examined on the HemoCue WBC DIFF, and the correlations between the instruments were assessed by Deming regression for total WBC, neutrophils, and lymphocytes. The theoretical CV% (CVt), calculated from number of cells counted by the HemoCue WBC DIFF, was used to determine the statistical error of the WBC counts. The interference of nRBC was also evaluated. RESULTS The counting variation was primarily the source of statistical error in the lower counts with an imprecision between 3.8-9.2% for total WBC (0.56-2.29 ×109 /L), 8.7-14.3% for neutrophils (0.36-1.33 ×109 /L) and 9.8-15.1% for lymphocytes (0.
