Through biological experiments, we found that it can complete cell imaging in macrophages and imaging of living mice, which has application prospects in Bioimaging. In addition, the probe NIRF has good applicability for Zn2+ and CN- detection in actual samples. The interactions between calf thymus DNA and a series of glucose-based cationic gemini surfactants 1a-1c with different spacer length, n = 4, 6 and 8, were studied by UV absorption, fluorescence spectroscopy, circular dichroism, FT-IR, dynamic light scattering and zeta potential measurements. The results showed that all the surfactants could interact with DNA efficiently. On addition of increasing concentration of the surfactants, UV absorption hypochromicity with insignificant blue shift were observed, until the DNA signal disappeared. The surfactant 1c was more efficient in the reduction of absorption intensity of DNA. According to the fluorescence quenching experiments by ethidium bromide exclusion, 1c exhibited the highest binding properties, with the binding constant at 3.25 × 108 L·mol-1. The spectroscopy study indicated that the surfactants bound with the DNA by a non-intercalative mode, mainly electrostatic interaction between the positively charged headgroups of the surfactants and negatively charged phosphate groups of DNA at low concentration, and the hydrophobic interaction among the alkyl chains at high concentration. The conformation of DNA during the interaction process could be kept B-form of DNA. For 1c, the DNA molecules can be compacted to about 103 nm in hydrodynamic diameter at 0.2 mM, while the minimum sizes of DNA were 140 nm and 133 nm, respectively, in the presence of 1a and 1b. The impact of the cationic gemini surfactants on the DNA compaction and condensation would shed light on their potential applications in gene delivery.Inflammation is a significant protective response in biological systems and associated with various diseases. Peroxynitrite (ONOO-) as a highly active oxidant participates in the inflammatory process of organisms. Thus, it is necessary to construct novel fluorescent probes for exploring inflammation-related diseases through detecting endogenous ONOO-. Resorufin-based fluorescent probes for testing ONOO- were rare and suffered from poor water solubility. In this work, we elaborately designed three resorufin-based incorporating isatin derivatives probes RF-ITs and successfully obtained two highly selective probes RF-IT-OC and RF-IT-EG for ONOO-. Comparing the other two probes, RF-IT-EG containing triethylene glycol monomethyl ether on isatin moiety displayed better water solubility (3.2 mg/L), faster response rate (60 s), larger signal-to-noise ratio (103-fold) and lower detection limit (87 nM) for monitoring ONOO-. The cells imaging results manifested that probe RF-IT-EG could be applied to trace endogenous ONOO- with inappreciable cytotoxicity. Moreover, the RF-IT-EG was capable of tracking the fluctuation of endogenous ONOO- in LPS-stimulated inflamed mouse leg models. This work will provide a faithful and promising probe for illustrating the roles of ONOO- in various inflammation-related diseases.Surface-enhanced Raman spectroscopy (SERS) is considered as an ultrasensitive, non-invasive as well as rapid detection technology for cancer diagnosis. In this study, we developed a novel blood serum analysis strategy using coffee ring effect-assisted label-free SERS for different types of cancer screening. Additionally, the pretreated Ag nanoparticles (Ag NPs) were mixed with the serum from liver cancer patients (n = 40), prostate cancer patients (n = 32) and healthy volunteers (n = 30) for SERS measurement. The droplets of Ag NPs-serum mixture formed the coffee ring on the peripheral after air-drying, and thus extremely enhancing Raman signal and ensuring the stability and reliability of SERS detection. Partial least square (PLS) and support vector machine (SVM) algorithms were utilized to establish the diagnosis model for SERS spectra data classifying, yielding the high diagnostic accuracy of 98.04% for normal group and two types of cancers simultaneously distinguishing. learn more More importantly, for the unknown testing set, an ideal diagnostic accuracy of 100% could be achieved by PLS-SVM algorithm for differentiating cancers from the normal group. link2 The results from this exploratory work demonstrate that serum SERS detection combined with PLS-SVM diagnostic algorithm and coffee ring effect has great potential for the noninvasive and label-free detection of cancer. We present complex spectroscopic data of the synthetic brunogeierite (Fe2+)2Ge4+O4 with space group Fd3¯m of spinel structure a = 8.3783 (6) Å, V = 588.12 (13) Å3. Brunogeierite crystals up to 200 μm in size were crystallized by the interaction of a boric acid solution on a metal iron wire in the presence of germanium oxide (GeO2) at 600/650 °C and 100 MPa. Mössbauer spectrum of synthetic brunogeierite consists of the symmetric doublet with the parameters IS = 1.104(1) mm/s and QS = 2.845(1) mm/s, that corresponds to the octahedral position of iron ions ([VI]Fe2+). The Raman spectra of Fe2GeO4 crystal consist of an intense main band at 756 cm-1 and four less intense bands at ∼644, 472, 302, and ∼205 cm-1 at ambient conditions. All five bands inherent in the spectrum of cubic spinel are present and gradual change in high pressure spectra up to 30 GPa. The color of the crystal changes from brown-orange to reddish at the center at 22.7 GPa and became opaque black up to 30.2 GPa. Herewith, in the high pressure spectra, we observed the splitting of some bands and the appearance of additional bands in a wide pressure range (from 1.6 to 30 GPa). The factor group analysis with the lattice dynamics calculation of potential crystal structure distortions shows the decreasing of the structure symmetry to tetragonal or rhombohedral in this pressure range.A compact gas sensing system based on the quartz crystal tuning fork (QCTF) and 2f/1f wavelength modulation spectroscopy (2f/1f-WMS) technique was reported for the first time. link3 An ingenious laser modulation strategy and frequency division multiplexing demodulation algorithm were developed for realizing a single QCTF to detect the first harmonic and second harmonic signals simultaneously. The influence of laser power change and excitation position the QCTF based 2f/1f-WMS technique was first investigated in detail. The results show that, compared with the traditional QCTF-2f method, the reported QCTF based 2f/1f technique has better immunity. To further evaluate this sensing technique, real-time monitoring of ambient water vapor (H2O) was made, the results show that the developed QCTF based 2f/1f-WMS technique can effectively improve the long-term stability and has super anti-interference ability to various environmental disturbance factors, such as light beam jitter, airflow fluctuation, and mechanical vibration, which proves it has great potential in practical field applications, especially for harsh environment.Metal ions released from metallic implants can affect the conformation and structural stability of proteins in biological fluids, which eventually affects the biocompatibility of implants. The present study aimed at understanding the interactions between the metal ions (Mn2+, Fe2+, Fe3+, Co2+, Cu2+, and Zn2+) and bovine serum albumin (BSA) molecules in physiological context. The structural information of BSA molecules and the microenvironment of functional groups were investigated using UV, Raman, and circular dichroism spectroscopy. The results revealed that addition of Fe3+, Fe2+, and Cu2+ ions alters the tertiary structure of BSA molecules and exposes the aromatic heterocyclic hydrophobic group of BSA amino acid residues. The addition of Fe3+ and Cu2+ ions results in increased viscosity and decreased intensity of the water peak in the BSA solution. Furthermore, Fe3+ and Cu2+ ions evidently promote the α-helix to β-sheet transformation of BSA molecules due to decreased disulfide bond stability. Tryptophan residues of BSA and metal ions containing BSA (Me+/BSA) solutions were found to be in a hydrophilic environment. Moreover, the addition of metal ions to BSA results in several of tyrosine residues acting as hydrogen-bond donors. Coomassie brilliant blue staining revealed that the addition of Cu2+ ions promotes the aggregation of BSA molecules. The findings of this study will be helpful for evaluating the biocompatibility of metallic implants.Nanopillars have been extensively explored as promising substrates for surface-enhanced Raman scattering (SERS) owing to their high sensitivity and excellent reproducibility. Most of the researches have been focused on the fabrication methods of nanopillars, and the dependences of SERS effects on geometrical size and spatial order are rarely investigated. In this work, SERS properties of nanopillars with different sizes (115-185 nm) and spatial orders (square and rhombus orders) have been studied. The work has shown that the nanopillars not only have high enhancement capability and high signal reproducibility, but also the enhancement is insensitive to the size and spatial orders. The measured enhancement factors (EFs) are 2.3-4.0 × 106 and signal reproducibility (relative standard deviation, RSD) are ∼ 5.2%-6.9%, which are among the best of the similar SERS substrates reported. The variation of SERS intensity was as low as approximately 4.8% with the variation of pillar size from 115, 135, 145, to 160 nm. The insensitiveness and high reproducibility have been ascribed to the combined excitation of localized surface plasmon resonance (LSPR) and propagating surface plasmons (SPPs) of the nanopillars. Optical properties of the nanopillars are studied both experimentally and numerically to understand the physics behind the SERS performance.Herein, a fluorescent nanosensor has been constructed for detection of glutathione (GSH) based on NH2-UiO-66 and AgNPs. The NH2-UiO-66 was synthesized where 2-amino-terephthalic acid as the organic ligand and Zr4+ as the center metal ions. The AgNPs can enhance the fluorescence of NH2-UiO-66 based on metal enhanced fluorescence (MEF) effect. Moreover, in the present of GSH, the fluorescence of NH2-UiO-66@AgNPs was quenched via electrostatic interaction and Ag-S reaction. The present sensing strategy shows good linear relation with the concentration of GSH in the range of 0.2-1.0 μM and 1.0-30 μM, and the limit of detection is 79 nM. Furthermore, our fluorescent nanosensor was utilized to detect GSH in human serum with a recovery of 96.8-102.5%. The results indicated that NH2-UiO-66@AgNPs is successfully applied for high sensitive and selective detection of GSH in human serum.Cabotegravir (CAB) is an antiretroviral therapy (ARV) used for Human Immunodeficiency Virus (HIV) treatment. CAB has low solubility, which affects its bioavailability in oral therapy. Moreover, the injection form of CAB has difficulty in the administration process. Therefore, it is essential to develop a new drug delivery system for CAB. Vaginal drug delivery system offers many advantages such as a large surface area, increased drug bioavailability, and improved drug delivery. CAB was developed in thermosensitive and mucoadhesive vaginal gel preparations that provided optimal distribution in the vaginal mucosa. To support the process of formulation development, in this study, UV-visible spectrophotometry method was validated in methanol, simulated vaginal fluid (SVF) and vaginal tissue to quantify the amount of CAB in the gel preparations, in vitro, and ex vivo studies, respectively. The developed analytical method was subsequently validated according to ICH guidelines. The calibration curves in these matrices were found to be linear with correlation coefficient values (R2) ≥ 0.
