The antitumor activity of the tea tree oil (TTO) derived product, Melaleuca Alternifolia Concentrate (MAC) was characterized mechanistically at the molecular and cellular level. MAC was analyzed for its anticancer activity against human prostate (LNCaP) and breast (MCF-7) cancer cell lines growing in vitro. see more MAC (0.02-0.06% v/v) dose-dependently induced the intrinsic (mitochondrial) apoptotic pathway in both the LNCaP and MCF-7 cell lines, involving increased mitochondrial superoxide production, loss of mitochondrial membrane potential (MMP), caspase 3/7 activation, as well as the presence of TUNEL+ and cleaved-PARP+ cell populations. At concentrations of 0.01-0.04% v/v, MAC caused cell cycle arrest in the G0/1-phase, as well as autophagy. The in vivo anticancer actions of MAC were examined as a treatment in the FVB/N c-Neu murine model for spontaneously arising breast cancers. Intratumoral MAC injections (1-4% v/v) significantly suppressed tumor progression in a dose-dependent manner and was associated with greater levels of tumor infiltrating neutrophils exhibiting anticancer cytotoxic activity. Induction of breast cancer cell death by MAC via the mitochondrial apoptotic pathway was also replicated occurring in tumors treated in vivo. In conclusion, our data highlights the potential for the Melaleuca-derived MAC product inducing anticancer neutrophil influx, supporting its application as a novel therapeutic agent.Our study has renewed interest in the genus Jasmine for the treatment of chronic inflammatory conditions. link2 Aerial parts of Jasminum grandiflorum L. subsp. floribundum total methanolic extract (JTME) were tested for its therapeutic potential as an anti-inflammatory agent using two experimental models in rats; acetic acid (AA) induced ulcerative colitis and adjuvant induced arthritis. The administration of JTME showed anti-inflammatory activity in a dose dependent manner. JTME, 400 mg/kg was like prednisolone, 2 mg/kg p.o. (the reference drug), since it improved the tissues of the colon clinically, macro and microscopically (ulcer index), and histopathological (scoring). It reduced the intestinal expression of pro-inflammatory cytokines in the colonic mucosa; IFNγ, TNFα, IL-6, IL-1, and MPO. It also preserved tight junctions in intestinal epithelial cells by counter-regulating claudin-5 and occludin levels additionally, it had a potent antioxidant activity. The expressions of NF-κB p65, TNF-α and caspase-3 in rats administered AA (2 mL of 4% solution, once, intrarectally) were significantly increased, where the lowest expression was scored in JTME, 400 mg/kg group. In the adjuvant induced model of rheumatoid arthritis, the TJME, 400 mg/kg reduced the levels of cathepsin D, iNOS, NO, RF, CRP, CPP and elevated the total antioxidant capacity of tissues. Additionally, it maintained bones without histopathological lesions, articular cartilage damage, and inflammation of the synovial membrane and periarticular tissues, in contrast to arthritic rats. Finally, we report a new detailed study to validate the medicinal importance of Jasminum for the chronic inflammatory disorders with immune dysfunction with anti-inflammatory and antioxidant effects.Numerous byproducts resulting from chlorinated disinfection are constantly being generated during water treatment processes. The potential risks of these new emerging pollutions remain largely unknown. Here, we determined the risks of chlorinated disinfection byproducts of diazepam (DZP) in the cellular and zebrafish exposure experiments. The cytotoxicity of disinfection byproducts (MACB and MBCC) was greater than DZP in macrophage raw 264.7 cells at 10 mg/L. We further found that the effects of MBCC on the metabolism of glycine, serine, threonine and riboflavin were far greater than DZP by the targeted metabolomics methods. Moreover, MBCC significantly decreased the peak amplitude of neuronal action potential in primary embryonic rat (Spragu-Dawley SD) hippocampal neurons. We finally determined behavioral toxicity of DZP and byproducts in zebrafish larvae. MBCC significantly decreased the maximal swim-activity and peak duration of zebrafish after 72 h exposure. Altogether, these findings indicate the MBCC pose serious pressures on public health.Boron (B) excess gives rise to a serious agricultural problem. In this study, we identified a B toxicity responsive transcription factor AtWRKY47 in Arabidopsis thaliana. The T-DNA insertion mutants Atwrky47 showed enhanced tolerance to B toxicity with better growth parameters under high B conditions compared to wild-type Col-0 plants. Quantitative analysis of AtWRKY47 mRNA abundance indicated that it was down-regulated under B toxicity conditions. Fluorescently labeled AtWRKY47 protein was localized in nucleus. In contrast to the phenotype of Atwrky47 mutants, overexpression of AtWRKY47 in Col-0 background resulted in lower biomass, less chlorophyll content, and increased sensitivity to B toxicity. More importantly, the B concentration in shoots was higher in the overexpression lines and lower in the Atwrky47 mutants than in Col-0 plants, respectively. These results demonstrate that AtWRKY47 gene plays a key role in regulating plant tolerance to B toxicity.2,4,6-trinitrotoluene (TNT) and cobalt (Co) contaminants have posed a severe environmental problem in many countries. Phytoremediation is an environmentally friendly technology for the remediation of these contaminants. However, the toxicity of TNT and cobalt limit the efficacy of phytoremediation application. The present research showed that expressing the Acidithiobacillus ferrooxidans single-strand DNA-binding protein gene (AfSSB) can improve the tolerance of Arabidopsis and tall fescue to TNT and cobalt. Compared to control plants, the AfSSB transformed Arabidopsis and tall fescue exhibited enhanced phytoremediation of TNT and cobalt separately contaminated soil and co-contaminated soil. The comet analysis revealed that the AfSSB transformed Arabidopsis suffer reduced DNA damage than control plants under TNT or cobalt exposure. In addition, the proteomic analysis revealed that AfSSB improves TNT and cobalt tolerance by strengthening the reactive superoxide (ROS) scavenging system and the detoxification system. Results presented here serve as strong theoretical support for the phytoremediation potential of organic and metal pollutants mediated by single-strand DNA-binding protein genes. SUMMARIZES This is the first report that AfSSB enhances phytoremediation of 2,4,6-trinitrotoluene and cobalt separately contaminated and co-contaminated soil.In aquaculture, fish are stressed with several factors involved in impacting the growth rate and health status. Although Nile tilapia can resist brackish water conditions, hypoxia status may impair the health condition of fish. Nile tilapia were exposed to salinity water at 0, 10, and 20‰ for four weeks then the growth behavior was checked. The results showed meaningfully lowered growth rate, feed utilization, and survival rate when fish kept in 20‰ for four weeks. Then fish were subdivided into six groups (factorial design, 2 × 3) in normoxia (DO, 6 mg/L) and hypoxia (DO, 1 mg/L) conditions for 24 h. High salinity (10 and 20‰) combined with hypoxia stress-induced inflammatory features in the intestines, gills, and livers of fish. The activities of SOD, CAT, and GPX were increased in the intestines, gills, and livers of fish grown in 10 and 20‰ and exposed with hypoxia stress. Fish grown in 20‰ and stressed with hypoxia had the highest ALT, AST, and ALP levels (p less then 0.05) among the groups. The highest transcription levels of Il-8, Il-1β, Ifn-γ, Tnf-α, and Caspase-3 genes and the lowest level of Il-10 gene were observed in fish exposed with 20‰ and hypoxia. The outputs of Integrated Biomarker Response (IBR) showed marked differences between fish groups with varied values. The lowest IBR was observed in fish reared in fresh water and normoxia, while the highest IBR was seen in the group of fish reared in 20‰ and hypoxia conditions (p less then 0.05). These results confirm that Nile tilapia can tolerate 10‰ in normoxia but 20‰ salinity combined with hypoxia results in oxidative stress, apoptosis, and inflammatory features in the intestines, gills, and livers. The obtained results indicate that hypoxia can affect the performances of Nile tilapia reared in brackish or high-water salinity leading to severe economic loss. Further future studies are required to understand the impact of different water salinities with hypoxia in the short term and long-term periods on the productivity of Nile tilapia.
To investigate the health-related quality of life (HRQOL) of Dutch adult childhood cancer survivors (CCS) and to identify risk factors of impaired HRQOL.
Adult CCS(age >18, diagnosed <18, ≥5 years since diagnosis) from the Dutch LATER registry completed the Medical Outcome Study Short Form 36 (SF-36) to measure HRQOL and provided sociodemographic characteristics. Age-adjusted mean SF-36 scale scores of CCS were compared to the Dutch general population for men and women separately using t-tests, with effect size d. Multivariate logistic regression models were built to identify sociodemographic and cancer-related risk factors for impaired physical and mental HRQOL.
Both male and female CCS (N=2301, mean age=35.4 years, 49.6% female) reported significantly (p≤.005) worse HRQOL than the general population on almost all scales of the SF-36 (-.11 ≤ d≤-.56). Largest differences were found on vitality and general health perceptions. Significant risk factors (p≤.05) for impaired physical HRQOL werefemale sex, older age at diagnosis, not having a partner, low educational attainment, disease recurrenceand exposure to radiotherapy, specifically to lower extremity radiation. Odds ratios (ORs) ranged from 1.6 to 3.7. Significant risk factors for impaired mental HRQOL wereage 26-35 years, male sex, not having a partnerand low educational attainment. ORs ranged from 1.3 to 2.0.
Adult CCS had worse HRQOL than the general population. CCS most at risk were those with low educational attainment and without a partner. Adult CCS could benefit from routine surveillance of their HRQOL. Special attention for CCS' vitality and health perceptions and beliefs is warranted.

Autism spectrum disorder is a common group of conditions affecting about one in 54 children. Electroencephalogram (EEG) signals from children with autism have a common morphological pattern which makes them distinguishable from normal EEG. link3 We have used this type of signal to design and implement an automated autism detection model.
We propose a hybrid lightweight deep feature extractor to obtain high classification performance. The system was designed and tested with a big EEG dataset that contained signals from autism patients and normal controls. (i) A new signal to image conversion model is presented in this paper. In this work, features are extracted from EEG signal using one-dimensional local binary pattern (1D_LBP) and the generated features are utilized as input of the short time Fourier transform (STFT) to generate spectrogram images. (ii) The deep features of the generated spectrogram images are extracted using a combination of pre-trained MobileNetV2, ShuffleNet, and SqueezeNet models. This method is named hybrid deep lightweight feature generator.
