Mg, Zn and Ce-doped CuO/HA composites were prepared by a two-step sol-gel and hydrothermal process. SEM images showed a spherical appearance of HA and a needle-like morphology for doped CuO. XRD patterns revealed that all doped CuO/HA composites exhibited a hexagonal crystal structure of HA and a monoclinic crystal structure of CuO with no impurities. ICP analysis indicated that with the increase of loading amount of doped CuO, the concentrations of Cu2+ ions and doping ions released from composites increased. Moreover, CuO/HA composites exhibit improved antibacterial properties against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) as compared with HA. When the loading amount of doped CuO in composites increased to 15 wt%, the composites exhibited the best antibacterial activity and complete bacterial growth inhibition effect. Furthermore, the CCK-8 assay revealed that the doped CuO/HA composites are noncytotoxic and can promote the proliferation of osteosarcoma cells. This work highlights the potential of the doped CuO/HA composites with significant antibacterial activity, bioactivity and cell compatibility for potential biomedical applications in dental implants and bone regeneration.Nanoparticles (NPs) can be produced via physical, chemical, or biological approaches. Yet, the impact of the synthesis approaches on the environmental fate and effects of NPs is poorly understood. Here, we synthesized AgNPs through chemical and biological approaches (cit-AgNPs and bio-AgNPs), characterized their properties, and toxicities relative to commercially available Ag nanopowder (np-AgNPs) to the clam Mercenaria mercenaria. The chemical synthesis is based on the reduction of ionic silver using sodium borohydride as a reducing agent and trisodium citrate as a capping agent. The biological synthesis is based on the reduction of ionic silver using biomolecules extracted from an atoxigenic strain of a filamentous fungus Aspergillus parasiticus. The properties of AgNPs were determined using UV-vis, dynamic light scattering, laser Doppler electrophoresis, (single particle)-inductively coupled plasma-mass spectroscopy, transmission electron microscopy, and asymmetric flow-field flow fractionation. Both chemiehaviors and should be considered for a more comprehensive environmental risk assessment of AgNPs.We report a general strategy to generate linear and circular gradients of active proteins or polymeric microparticles on planar surfaces by controlling the distribution of electrostatic field during electrohydrodynamic jet printing or electrospray process. Taking fibronectin as an example, we generated a circular gradient of fibronectin and investigated its effect on accelerating the migration of fibroblasts to suit for use in wound closure. In another demonstration, we created linear gradients of laminin in unidirectional and bidirectional patterns, respectively. We showed that such gradations significantly promoted the migration of human neuroblastoma cells with the increase of laminin content. When we changed fibronectin/laminin to electrosprayed poly(lactic-co-glycolic acid) (PLGA) microparticles, we found similar results in terms of guiding cell migration, except that the guidance cues varied from biological signal to topographic structure. Taken together, this method for generating linear/circular gradients of fibronectin/laminin and PLGA microparticles can be readily extended to different types of bioactive proteins and polymeric microparticles to suit wound closure, nerve repair, and related applications involving cell migration.This study examined the application of active inference to dynamic visuomotor control. Active inference proposes that actions are dynamically planned according to uncertainty about sensory information, prior expectations, and the environment, with motor adjustments serving to minimise future prediction errors. We investigated whether predictive gaze behaviours are indeed adjusted in this Bayes-optimal fashion during a virtual racquetball task. In this task, participants intercepted bouncing balls with varying levels of elasticity, under conditions of higher or lower environmental volatility. Participants' gaze patterns differed between stable and volatile conditions in a manner consistent with generative models of Bayes-optimal behaviour. Partially observable Markov models also revealed an increased rate of associative learning in response to unpredictable shifts in environmental probabilities, although there was no overall effect of volatility on this parameter. Findings extend active inference frameworks into complex and unconstrained visuomotor tasks and present important implications for a neurocomputational understanding of the visual guidance of action.Hydroxy-epoxy- and trihydroxy derivatives of linoleic acid are proposed to play an essential function in formation of the mammalian skin permeability barrier, which could account for the essential nature of its precursor, linoleic acid. Recent literature suggests that a specific oxidized enone derivative of LA esterified in ceramides facilitates binding to proteins, potentially serving a structural role in formation of the epidermal skin barrier. However, it is still to be established if other linoleic acid derivatives are also required for skin barrier formation, and whether the essential role is performed exclusively by an esterified, structural lipid or as an unesterified, labile signaling lipid, or by some combination of these derivatives. Progress in this domain is limited by lack of availability of hydroxy‑epoxy-and trihydroxy- and octadecenoate derivatives of linoleic acid and related compounds, and challenges in maintaining them in the unesterified lipid pool. Here we describe methods for the total synthesis of hydroxy‑epoxy-octadecenoate derivatives of linoleic acid (HEL1), and stable analogs that are designed to be resistant to inactivation by (a) acylation/esterification (thus trapping these lipids in the free acid pool), (b) dehydrogenation, and (c) analogs combining both modifications. We further provide a total synthesis of corresponding hydroxy‑epoxy- derivatives of sebaleic acid (a regioisomer of linoleic acid present in skin), and of small molecule scaffolds containing the allylic and non-allylic epoxide 7-carbon substructures shared by both families of hydroxy‑epoxy-and trihydroxy- octadecenoates. Finally, we demonstrate that 2,2-dimethyl analogs of hydroxy‑epoxy-and trihydroxy- octadecenoates are resistant to esterification with an in vitro assay and thus provide a novel template for stabilizing labile, bioactive lipids as free acids by preventing acylation/esterification.The ability of Salmonella to resist and adapt to harsh conditions is one of the major features that have made this microorganism such a relevant health hazard. However, the impact of these resistance responses on other aspects of Salmonella physiology, such as virulence and growth ability, is still not fully understood. The objective of this study was to determine the maximum growth rates (in three different media), virulence (adhesion and invasion of Caco-2 cells), and other phenotypic characteristics (biofilm-forming ability and antimicrobial resistance) of 23 Salmonella strains belonging to different serovars, and to compare them with their previously determined stress resistance parameters. Significant differences (p less then 0.05) in growth rates, virulence, and biofilm-forming ability were found among the 23 strains studied. Nevertheless, whereas less than 3-fold change between the lowest and the highest growth rate was observed, the percentage of cells capable of invading Caco-2 cells varied more tht are relevant from the food safety perspective.High levels of biogenic amines (BAs) in fermented food can present a health risk to consumers. Microorganisms that can reduce BAs are widely used in fermented foods. However, the mechanism by which microorganisms reduce BAs in foods has not been explored. In this study, we investigated how Staphylococcus nepalensis 5-5 (S. nepalensis 5-5), which was a BA-degrading strain isolated from fish sauce, could reduce BA accumulation in the fish sauce. High-throughput sequencing and HPLC methods were sequentially used to determine the microbial community structure and BA content in fish sauce with/without S. nepalensis 5-5. The results showed that S. nepalensis 5-5 might be a safe strain that could improve the flavor of fish sauce while still exhibiting good BA degradation ability under a high salt environment. The content of BAs in fish sauce inoculated with S. nepalensis 5-5 was significantly decreased compared with the control fish sauce, achieving maximal reductions of 15.74, 14.18 and 16.65% in putrescine, cadaverine and histamine accumulation, respectively. According to high-throughput sequencing data, S. nepalensis 5-5 reduced the abundance of the genera positively associated with BAs, while increasing the number of bacterial genera negatively correlated with BAs in the sample and changed the correlation between some genera and BAs via species interaction. In addition, analysis of amino acid metabolism showed that S. nepalensis 5-5 might use histidine to produce metabolites other than histamine, thereby reducing the production of BAs. These findings not only explained the mechanisms by which the BA level in fish sauce could be reduced but also provided a potential means to control BA production in the fish sauce during the fermentation stage."Torta del Casar" is a Spanish soft-ripened cheese made with sheep's raw milk and subjected to a short ripening process, which favors the growth of pathogenic microorganisms including Listeria monocytogenes. The development of strategies to control pathogens and minimize health risks associated with the presence of L. monocytogenes in these products is of great interest. AZD9291 In this regard, the anti-Listeria activity of a whey protein hydrolysate (ProH) alone or combined with six lactic acid bacteria strains isolated from cheese was evaluated in this study as a biocontrol strategy using a "Torta del Casar" cheese-based medium. The most active combinations of lactic acid bacteria assayed induced a reduction higher than two logarithmic units in the growth of L. monocytogenes (serotype 4b) compared to their respective control when they were co-inoculated in "Torta del Casar" cheese-based medium at 7 °C for 7 days. In addition, the observed downregulation of some key virulence genes of L. monocytogenes suggests that the strain Lactiplantibacillus plantarum B2 alone and combined with the strain Lactiplantibacillus spp. B4 are good candidates to be used as biocontrol agents against L. monocytogenes growth in traditional soft cheeses based on raw milk during their storage at refrigeration temperatures.A two-dimensional nitrogen-rich carbon nitrogen (C3N5) material was prepared via a facile high temperature thermal polymerization. For the first time, the C3N5 was used as fiber coating of solid-phase microextraction (SPME) to extract and preconcentrate polychlorinated biphenyls (PCBs) before gas chromatography (GC) analysis. The X-ray diffraction, N2 adsorption-desorption, Fourier transform-infrared spectroscopy, thermogravimetric analysis, and X-ray photoelectron spectroscopy were performed to investigate structure, functional groups, thermal stability, bonding type, element composition, and atomic ratio of C3N5. The two-dimensional planar stacking structure was further verified by scanning electron microscopy and transmission electron microscopy. Five PCBs including PCB-4, PCB-12, PCB-29, PCB-52 and PCB-101 were selected as targets to evaluate performance of SPME fiber. Under the optimal conditions, the method showed a good linear range from 0.01 to 1000 ng/mL with the correlation coefficients (R2) higher than 0.
