This review summarizes the role of MSC-EVs in normal and malignant hematopoiesis and their potential contribution in treating GVHD.The publication presents a comparison of the sound absorption test results of a perforated wall cassette filled with mineral wool for various degree of cement dust pollution. Cement dust should be understood here as dust created during the production of cement and during the milling and dispatch of finished products. If the partitions in production plants are made of sound-absorbing cassettes or additional sound-absorbing elements made of perforated cassettes are applied, we must know how dust can change sound-absorbing properties of the cassettes. Thus, one has to consider whether the use of sound-absorbing perforated cassettes is appropriate if sound-absorbing parameters change over time due to dust. To determine the impact of dust-covered perforation on sound-absorbing parameters, tests were performed for four variants having different level of pollution. The tests involved 'clean' and then dust-covered cassettes, each time increasing the amount of cement dust on the perforations. Sound absorption parameters of the cassettes were tested in the reverberation chamber for individual variants. Test results indicate the loss of sound absorption of the cassettes only when they are heavily polluted. Then the reduction of the single-number sound absorption index αw is 50%. Using computer simulation, we analyzed how the change of sound-absorbing parameters of the cassettes would influence the change of noise reduction in the production hall. The results of the analysis demonstrate a very effective reduction of noise level of 14 dB by the application of clean cassettes. The reduction value for the dirtiest cassettes was 6 dB.(1) One strategy to improve the outcome of orthopedic implants is to use porous implants with the addition of a coating with an antibacterial biomolecule. In this study, we aimed to produce and test the biocompatibility, the osteopromotive (both under normal conditions and under a bacterial challenge with lipopolysaccharide (LPS)) and antibacterial activities of a porous Ti-6Al-4V implant coated with the flavonoid quercitrin in vitro. (2) Porous Ti-6Al-4V implants were produced by 3D printing and further functionalized with quercitrin by wet chemistry. Implants were characterized in terms of porosity and mechanical testing, and the coating with quercitrin by fluorescence staining. Implant biocompatibility and bioactivity was tested using MC3T3-E1 preosteoblasts by analyzing cytotoxicity, cell adhesion, osteocalcin production, and alkaline phosphatase (ALP) activity under control and under bacterial challenging conditions using lipopolysaccharide (LPS). Finally, the antibacterial properties of the implants were studied using Staphylococcus epidermidis by measuring bacterial viability and adhesion. (3) Porous implants showed pore size of about 500 µm and a porosity of 52%. The coating was homogeneous over all the 3D surface and did not alter the mechanical properties of the Young modulus. Quercitrin-coated implants showed higher biocompatibility, cell adhesion, and osteocalcin production compared with control implants. Moreover, higher ALP activity was observed for the quercitrin group under both normal and bacterial challenging conditions. Finally, S. epidermidis live/dead ratio and adhesion after 4 h of incubation was lower on quercitrin implants compared with the control. (4) Quercitrin-functionalized porous Ti-6Al-4V implants present a great potential as an orthopedic porous implant that decreases bacterial adhesion and viability while promoting bone cell growth and differentiation.Cured-in-place pipe (CIPP), as a kind of trenchless sewer rehabilitation technology, is a method to repair sewer pipe using unsaturated polyester resin. This study develops a CIPP liner using hot water or steam curing as well as glass fiber, in contrast to traditional methods, which use nonwoven fabric. Composite material samples were fabricated by combining liner materials using various methods, and the structural characteristics of the liners were compared and analyzed through short- and long-term flexural strength tests. A long-term test was conducted for 10,000 h, and the results revealed 13.3 times higher flexural strength and 8 times higher flexural modulus than the American Society for Testing Materials minimum criteria for CIPP short-term properties. The maximum creep retention factor was 0.64, thereby reducing the design thickness of the CIPP by up to 54%. The structural characteristics also improved when glass fibers were mixed with traditional CIPP liner, making it possible to reduce the thickness by 30%. Glass fibers result in high structural strength when combined with unsaturated polyester resin. Structural strength increased, even when glass fibers were mixed with traditional CIPP liner. The main contribution of this research is the development of a high strength CIPP liner and improvement of the structural properties of CIPP lining without using the specially formulated resin or lining materials.New tools for activating endodontic irrigants have evolved, yet their impact on root canal disinfection, in comparison to the passive placing of an inter-visit medication, have not yet been fully elucidated. The use of DNA- and rRNA-based methods may cast some new light on this issue, as they allow a comparison to be made between microbial presence and activity. Therefore, the aim of this single-arm intervention trial is to evaluate the antibacterial effect of endodontic procedures using both molecular methods. Root canal samples were obtained from 20 patients with asymptomatic apical periodontitis after each treatment step access cavity, chemo-mechanical preparation, adjunctive procedures (XP-endo Finisher file and passive ultrasonic irrigation), calcium hydroxide medication, and 2nd-visit root canal preparation. DNA and cDNA from the samples were subjected to quantitative polymerase chain reaction with universal primers for the bacterial 16S rRNA gene. Chemo-mechanical preparation promoted a drastic reduction in bacterial levels and activity, whereas the adjunctive procedures did not make a significant contribution to further disinfection. At the 2nd visit, bacteria were active after the use of calcium hydroxide medication; however, they were significantly reduced after a 2nd-visit preparation. Consequently, the lowest bacterial levels were found at the end of the treatment. This clinical trial, which used an rRNA and rDNA combined approach, confirmed previous studies showing that root canal preparation represents the main strategy for root canal disinfection.Trials using antisense oligonucleotide technology to lower Huntingtin levels in Huntington's disease (HD) are currently ongoing. This progress, taking place only 27 years after the identification of the Huntingtin gene (HTT) in 1993 reflects the enormous development in genetic engineering in the last decades. It is also the result of passionate basic scientific work and large worldwide registry studies that have advanced the understanding of HD. Increased knowledge of the pathophysiology of this autosomal dominantly inherited CAG-repeat expansion mediated neurodegenerative disease has led to the development of several putative treatment strategies, currently under investigation. These strategies span the whole spectrum of potential targets from genome editing via RNA interference to promoting protein degradation. Yet, recent studies revealed the importance of huntingtin RNA in the pathogenesis of the disease. Therefore, huntingtin-lowering by means of RNA interference appears to be a particular promising strategy. As a matter of fact, these approaches have entered, or are on the verge of entering, the clinical trial period. Here, we provide an overview of huntingtin-lowering approaches via DNA or RNA interference in present clinical trials as well as strategies subject to upcoming therapeutic options. We furthermore discuss putative implications for future treatment of HD patients.The Western diet can lead to alterations in cardiac function and increase cardiovascular risk, which can be reproduced in animal models by implementing a high-fat diet (HFD). However, the mechanism of these alterations is not fully understood and may be dependent on alterations in heart lipid composition. The aim of this study was to evaluate the effect of an HFD on the fatty acid (FA) composition of total lipids, as well as of various lipid fractions in the heart, and on heart function. C57BL/6 mice were fed an HFD or standard laboratory diet. The FA composition of chow, serum, heart and skeletal muscle tissues was measured by gas chromatography-mass spectrometry. Cardiac function was evaluated by ultrasonography. Our results showed an unexpected increase in polyunsaturated FAs (PUFAs) and a significant decrease in monounsaturated FAs (MUFAs) in the heart tissue of mice fed the HFD. For comparison, no such effects were observed in skeletal muscle or serum samples. Furthermore, we found that the largest increase in PUFAs was in the sphingolipid fraction, whereas the largest decrease in MUFAs was in the phospholipid and sphingomyelin fractions. The hearts of mice fed an HFD had an increased content of triacylglycerols. Moreover, the HFD treatment altered aortic flow pattern. We did not find significant changes in heart mass or oxidative stress markers between mice fed the HFD and standard diet. The above results suggest that alterations in FA composition in the heart may contribute to deterioration of heart function. A possible mechanism of this phenomenon is the alteration of sphingolipids and phospholipids in the fatty acid profile, which may change the physical properties of these lipids. Since phospho- and sphingolipids are the major components of cell membranes, alterations in their structures in heart cells can result in changes in cell membrane properties.Heteroptera, or true bugs populate many climate zones, coping with different environmental conditions. The aim of this study was the evaluation of their thermal limits and derived traits, as well as climatological parameters which might influence their distribution. We assessed the thermal limits (critical thermal maxima, CTmax, and minima, CTmin) of eight seed bug species (Lygaeidae, Pyrrhocoridae) distributed over four Köppen-Geiger climate classification types (KCC), approximately 6° of latitude, and four European countries (Austria, Italy, Croatia, Bulgaria). In test tubes, a temperature ramp was driven down to -5 °C for CTmin and up to 50 °C for CTmax (0.25 °C/min) until the bugs' voluntary, coordinated movement stopped. In contrast to CTmin, CTmax depended significantly on KCC, species, and body mass. CTmax showed high correlation with bioclimatic parameters such as annual mean temperature and mean maximum temperature of warmest month (BIO5), as well as three parameters representing temperature variability. CTmin correlated with mean annual temperature, mean minimum temperature of coldest month (BIO6), and two parameters representing variability. GF109203X Although the derived trait cold tolerance (TC = BIO6 - CTmin) depended on several bioclimatic variables, heat tolerance (TH = CTmax - BIO5) showed no correlation. Seed bugs seem to have potential for further range shifts in the face of global warming.
