Loss of KDM6A was associated with metastasis in PDAC patients. Bru-seq analysis revealed upregulation of the epithelial-mesenchymal transition pathway in PDAC cells deficient of KDM6A. Loss of KDM6A promoted mesenchymal morphology, migration, and invasion in PDAC cells in vitro. Mechanistically, activin A and subsequent p38 activation likely mediated the role of KDM6A loss. Inhibiting either activin A or p38 reversed the effect. Pancreas-specific Kdm6a-knockout mice pancreata demonstrated accelerated PDAC progression, developed a more aggressive undifferentiated type PDAC, and increased metastases in the background of Kras and p53 mutations. Kdm6a-deficient pancreata in a pancreatitis model had a delayed recovery with increased PDAC precursor lesions compared to wild-type pancreata.
Loss of KDM6A accelerates PDAC progression and metastasis, most likely by a non-canonical p38-dependant activin A pathway. KDM6A also promotes pancreatic tissue recovery from pancreatitis. Activin A might be utilized as a therapeutic target for KDM6A-deficient PDACs.

Activin A might be utilized as a therapeutic target for KDM6A-deficient PDACs.Our aim was to assess the incidence of symptomatic ulnar nerve dislocation and its influence on surgical outcome after primary and revision surgeries in ulnar nerve entrapment at the elbow (ulnar neuropathy at the elbow (UNE) or cubital tunnel syndrome). MGCD0103 The influence of pre- or intra-operative ulnar nerve dislocation on postoperative outcome was assessed in 548 surgically treated cases (548 nerves) from two hand surgery departments reporting to the Swedish National Quality Registry for Hand Surgery, using QuickDASH, a patient-reported outcome measure (PROM), before surgery and at 3 and 12 months postoperatively, and a doctor-reported outcome measure (DROM), grading as "cured-improved "or "unchanged-worsened," at a median follow-up of 3.0 months [IQR, 1.5-6.0]. 109 of the 548 cases (20%) showed documented pre- or intra-operative ulnar nerve dislocation; more often found at revision (35/75, 47%) than at primary surgery (74/473, 16%) (p less then 0.0001). Cases with dislocation presented higher QuickDASH scores at 12 months (p = 0.026). A linear regression model, adjusted for age and gender, predicted higher QuickDASH scores at 12 months postoperatively for cases with dislocation (unstandardized B 11.3 [95% CI 0.4-22.2], p = 0.043). DROM grading as unchanged-worsened at a median 3 months predicted worse QuickDASH scores (p less then 0.0001) than in cured-improved cases at 3 (unstandardized B, 18.4 [95% CI 9.4-27.3]) and 12 months (unstandardized B, 18.1 [9.1-27.0]). Primary surgeries had better DROM grading than revision surgeries (p = 0.033; cured-improved, 75% and 63%, respectively), but QuickDASH scores did not differ. Presence of a clinically relevant ulnar nerve dislocation resulted in worse outcome, perhaps due to more extensive surgery with transposition. Nerve dislocation needs attention when treating UNE patients.The Stroop task, which examines an aspect of executive function/cognitive control, the ability to inhibit prepotent responses, has been relatively little examined in schizophrenia, and the findings have been inconsistent. Whether performance of this task is associated with failure of de-activation in the disorder is also uncertain. We examined 42 schizophrenic patients and 61 healthy controls during performance of an fMRI-adapted version of the Stroop task, the counting Stroop task. Task-related activations (incongruent > congruent condition) and de-activations (baseline > incongruent) were examined using whole-brain, voxel-based methods. In the healthy controls, task performance was found to be associated with activations in the left dorsolateral prefrontal cortex and the dorsal anterior cingulate cortex, among other regions. De-activations were seen in the medial frontal cortex, the middle and posterior cingulate gyrus and cuneus, the parahippocampal gyrus and the hippocampus. The schizophrenic patients did not show reduced activation compared to the healthy controls. They did, however, show failure of de-activation in the medial frontal cortex. Our negative finding with respect to hypoactivation during performance of a task requiring inhibition of prepotent responses suggests that brain functional abnormality in schizophrenia may not affect all aspects of executive function/cognitive control. The finding of medial frontal cortex failure of de-activation adds to existing findings of default mode network dysfunction in the disorder.Atmospheric dispersion models are crucial for nuclear risk assessment and emergency response systems since they rapidly predict air concentrations and deposition of released radionuclides, providing a basis for dose estimations and countermeasure strategies. Atmospheric dispersion models are associated with relatively large and often unknown uncertainties that are mostly attributed to meteorology, source terms and parametrisation of the dispersion model. By developing methods that can provide reliable uncertainty ranges for model outputs, decision makers have an improved basis for handling nuclear emergency situations. In the present work, model skill of the Severe Nuclear Accident Programme (SNAP) model was quantified by employing an ensemble method in which 51 meteorological realisations from a numerical weather prediction model were combined with 9 source term descriptions for the accidental 137Cs releases from Fukushima Daiichi Nuclear Power Plant during 14th-17th March 2011. The meteorological forecast wcident, which emphasises the importance of including ensemble prediction in nuclear preparedness tools of the future.Phthalates (PAEs) accumulated in agricultural soils and rice have increased human exposure risks. Microbial degradation could efficiently reduce the residue of organic pollutants in soil and crop plants. Here, we hypothesized that endophytic bacteria from wild rice have the potential for degradation of PAEs and plant growth promoting. The endophytic bacterial community and functional diversity in wild rice (Oryza meridionalis) were analyzed for the first time, and the potential for PAE degradation and plant growth promoting by endophytes were investigated. The results of Illumina high-throughput sequencing revealed that abundant endophytes inhabited in wild rice with Proteobacteria, Bacteroidetes, Firmicutes and Actinobacteria being the dominant phyla. Endophytic bacterial diversity and complexity were confirmed by isolation and clustering of isolates. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis demonstrated that endophytes exerted diverse functions such as plant growth promoting, xenobiotics biodegradation, pollution remediation and bacterial chemotaxis. Pure culture experiment showed that 30 isolated endophytic strains exhibited in vitro plant growth promoting activities, and rice plants inoculated with these strains confirmed their growth promoting abilities. Some endophytic strains were capable of efficiently degrading PAEs, with the highest removal percentage of di-n-butyl phthalate (DBP) up to 96.1% by Bacillus amyloliquefaciens strain L381 within 5 days. Synthetic community F and strain L381 rapidly removed DBP from soil (removing 91.0%-99.2% within 10 d and from rice plant slurry (removing 93.4%-99.2% within 5 d). These results confirmed the hypothesis and demonstrated the diversity of endophytic bacteria in wild rice with diverse functions, especially for plant growth promoting and removing PAEs. These multifunctional endophytic bacteria provided good alternatives to reduce PAE accumulation in crops and increase yield.Excessive ammonia-nitrogen (N) in aquifers has caused groundwater pollution on the Dongting Plain (DTP), which seriously threatens the safety of drinking water. It is urgent and necessary to determine the sources and enrichment mechanisms of ammonia-N in groundwater. Carrying out water and soil collaborative research on a three-dimensional scale can more comprehensively analyze the sources of N, including natural and anthropogenic sources. In this study, surface and groundwater quality characteristics were examined from a total of 77 sites on the DTP. Two subsequent boreholes were drilled in the high ammonia concentration area and normal groundwater area, respectively, to compare the effects of anthropogenic activities. Indicators from hydrogeochemical and pedogeochemical analyses, as well as various isotopes, including δ15N-NH4+, δ15N-TON, δ15N-NO3-, δ18O-NO3-, δ18O-H2O, δD-H2O, and δ13C-TOC were used to identify ammonium sources and transformation mechanisms in the strata. We found that the sediments were cents and ammonia in groundwater.Aging is an inevitable natural process, leading to faded performances of soil amendments. Understanding long-term aging features is crucial for the risk management of contaminated soil. In this study, a novel quantitative aging method, namely, the "soil coin" method, was developed, which can simulate the effects of natural aging on metal(loid) immobilization performances. To better depict the aging features, two models on the basis of conditional probability-induced failure were developed. To effectively immobilize soil arsenic (As) and antimony (Sb), magnesium (Mg) and iron (Fe) oxides were simultaneously introduced to either fresh or pre-oxidized biochar via a facile method. Although post-application aging is harmful, pre-aging (i.e., pre-oxidation using H2O2) in turn served as an effective means to introduce more metal oxides, thereby rendering better short-term and long-term effectiveness for metalloid immobilization. Experimental and modeling approaches suggested that precipitation accounted for long-term immobilization, while a constant aging rate is the key feature for a promising soil amendment. It is suggested that to further calibrate this method and better understand the immobilization performances in the long run, more evidence from the field is needed.In this study, for the first time, we developed a series of co-graft tannin-based flocculants, TA-g-P(AM-DMDAAC), with different charge densities (CDs) and molecular weights (MWs) and evaluated their algal-removal performances. The effects of TA-g-P(AM-DMDAAC) on the cell integrity of Microcystis aeruginosa and release of extracellular organic matter (EOM) and microcystin-leucine-arginine (MC-LR) in flocculation and floc storage were also studied. Results suggested that TA-g-P(AM-DMDAAC) could not only efficiently remove algal cells over a wide pH range (pH 3-11) but also EOM. CD and MW significantly affected flocculation performance and floc characteristics of TA-g-P(AM-DMDAAC). A higher CD helped achieve a higher removal efficiency of algal cells and EOM, whereas a higher MW resulted in the formation of larger and more compact flocs. Furthermore, the larger and denser flocs could better protect algal cells and reduce the release of EOM during floc storage. Notably, algal cells in the TA-g-P(AM-DMDAAC) flocs did not appear to show signs of massive rupture nor did they release EOM and MC-LR extensively for at least 20 days of storage.
