To investigate the clinical response to infliximab in ocular inflammation patients who develop anti-infliximab antibodies (AIA) vs. those patients who do not develop AIA.

A retrospective review was performed of patients treated with infliximab for noninfectious uveitis (NIU) or scleritis. Clinical response was determined as a composite clinical endpoint and classified as complete, partial, or absent. Nine of 32 infliximab-treated patients (28%) were found to develop AIA. Among the AIA-positive patients, clinical response was complete in 7 patients (78%) and partial in 2 patients (22%). Among the AIA-negative patients, clinical response was complete in 15 patients (65%), partial in 6 patients (26%) and absent in 2 patients (9%). Serum infliximab levels tended to decrease with appearance of AIA but rarely became undetectable.

In this pilot study, AIA-positive patients did not have diminished clinical response to infliximab when compared with AIA-negative patients. There was a high rate of complete clinical response to infliximab in this group of NIU and scleritis patients. Approximately a quarter of patients developed AIA. AIA-positive patients did not have diminished rates of clinical response when compared with AIA-negative patients. This suggests that routine AIA monitoring may not be clinically useful, although validation of this finding in larger cohorts is necessary.

As a result, the aims of the present study were to screen secondary metabolites and to evaluate in vitro antifungal activities of Brucea antidysenterica, Aloe vera, and Justicia schimperiana. After the plants were identified, their leaves were collected, washed, dried under the shade, pulverized, and extracted with methanol (99.8%) using the maceration technique. The presence of secondary metabolites in plant extracts was screened using various laboratory protocols. WM-8014 concentration The antifungal activities of the plant extract against reference fungal strains of Candida albicans and Aspergillus niger at concentrations of 200, 100, and 50 mg/mL were assessed using the agar-well diffusion method. Ketoconazole (15 μg) was used as a positive control, while 5% dimethyl sulfoxide and/or 5% Tween 80 were used as negative controls. All tests were conducted in triplicate. Alkaloids, flavonoids, and phenols were secondary metabolites found in all plant extracts. The extract of leaves of B. antidysenterica and J. schimperiana formed a mean zone of inhibition of 15.5 ± 0.5 mm and 15.3 ± 0.58 mm, respectively, against Candida albicans at a concentration of 200 mg/mL, whereas extracts of A. vera leaves formed a 12.3 ± 0.58 mm inhibition zone only against Aspergillus niger at 200 mg/mL. In conclusion, the current study found that B. antidysenterica, A. vera, and J. schimperiana had antifungal activity. In addition, all these plants had a variety of secondary metabolites that possibly have antifungal activities. Studies on in vivo investigations and isolation of specific antifungal compounds from these medicinal plants are suggested.Metasurfaces consisting of artificially designed meta-atoms have been popularized recently due to their advantages of amplitude and phase of light control. However, the electron beam lithography method for metasurface fabrication has high cost and low throughput, which results in a limitation for the fabrication of metasurfaces. In this study, nanocomposite printing technology is used to fabricate high-efficiency metasurfaces with low cost. To demonstrate the efficiency of the proposed fabrication method, a metahologram is designed and fabricated using a nanocomposite. The metahologram exhibits conversion efficiencies of 48% and 35% at wavelengths of 532 and 635 nm, respectively. The nanocomposite is composed of polymers with nanoparticles, so durability tests are also performed to evaluate the effects of temperature and humidity on the metasurfaces. The test verifies that at temperatures below the glass transition temperature of the base resin, the nanostructures do not collapse, so the efficiency of the metasurfaces remains almost the same. The surrounding humidity does not affect the nanostructures at all. Hence, the durability of the nanocomposite metasurfaces can be further enhanced by replacing the base resin, and this nanocomposite printing method will facilitate practical metasurface use at low cost.Hypothenar hammer syndrome is a rare cause of vascular insufficiency. Generally, patients report a history of repetitive trauma to the hypothenar region of the hand. Symptoms often consist of cold intolerance, pain, paleness, and paresthesia due to digital ischemia. The severity of these symptoms will depend on the extent of ulnar artery occlusion and the presence or absence of collaterals between this artery's superficial and deep branches. It is a rare clinical entity, which on multiple occasions requires a surgical approach. We present a 63-year-old man with bilateral Raynaud's phenomenon secondary to hypothenar hammer syndrome successfully treated by vascular repair surgery. In patients with Raynaud's phenomenon, it is important to know that there are reversible causes such as hypothenar hammer syndrome.The effects of external factors such as temperature, humidity, pesticide formulation, and pesticide concentration on the contact angle of pesticide droplets on rice leaf surfaces were analyzed. The experiments showed that there were significant differences in the contact angles of droplets on the leaf surfaces under different temperatures and humidity. As the ambient temperature increased, the contact angle first decreased and then increased, reaching a minimum value at 25°C. With a gradual increase in humidity, the contact angle significantly increased and reached a maximum at 100% humidity. Finally, it was concluded that both the formulation and concentration of the pesticide had a significant effect on the contact angle of droplets on rice leaf surfaces. The experiments also illustrated that the effects of the pesticide formulation and concentration on the contact angle were more significant than those of temperature and humidity.The ability to predict the environmental behavior of chemicals precisely is important for realizing more rational regulation. In this study, the bioaccumulation of nine chemicals of different molecular weights absorbed via the intestinal tract was evaluated in fish using the everted gut sac method. The amounts of chemicals that passed through the intestinal membrane after a 24-hr exposure were significantly decreased for chemicals with MW≥548 and Dmax min≥15.8 Å (or Dmax aver≥17.2 Å). These thresholds are consistent with those previously proposed in terms of MW (>800) and molecular size (Dmax min>15.6 Å or Dmax aver>17.1 Å) for the limit of permeable chemicals through the gill membrane. The results show that the same MW and Dmax criteria can be used to predict low bioaccumulation through both the gill membrane and the intestinal tract. These findings are helpful in reducing the need to conduct animal tests in environmental safety studies.γ-Aminobutyric acid receptors (GABARs) mediate fast inhibitory neurotransmission and are targets for insecticides. GABARs are composed of five subunits, the composition of which dictates the pharmacological characteristics of GABARs. Both competitive and noncompetitive GABAR antagonists can be used as insecticides. Gabazine is a potent competitive antagonist of mammalian α1β2γ2 GABARs; however, it is less potent against insect GABARs. To explore how gabazine interacts with GABARs, we examined whether the sensitivity of the small brown planthopper (Laodelphax striatellus) RDL GABAR (LsRDLR) to gabazine is increased when its amino acid residues are substituted with α1β2γ2 GABAR residues. In the results, two of the generated mutants showed enhanced gabazine sensitivity. Docking simulations of gabazine using LsRDLR homology models and an α1β2γ2 GABAR cryo-EM structure revealed that the accommodation of gabazine into the "aromatic box" in the orthosteric site lowered the binding energy. This information may help in designing GABAR-targeting insecticides with novel modes of action.Orius insidiosus, known as the pirate bug, is widely distributed throughout the Americas. It is employed for the biological control of Frankliniella occidentalis in organic berry crops in Mexico. In conventional crops, spinosad is the main control method for this pest. The LD50 of spinosad on O. insidiosus was determined. In addition, we monitored the population density of F. occidentalis in blackberry crops under two types of management (biochemical+mass trapping, and biological control). The LD50 was 225.65 ppm 3.8 times greater than the 60 ppm dose commonly used in blackberry crops. Both types of control are efficient; however, spinosad is less effective and should be combined with other environmentally friendly strategies. The possibility of combining chromatic traps+spinosad application and chromatic traps+strategic release of O. insidiosus to effectively control thrips without compromising fruit quality is discussed.Bacillus thuringiensis (Bt) has been used as sprayable pesticides for many decades. Bt strains utilized in these products produce multiple insecticidal proteins to complement a narrow insect specificity of each protein. In the late 1990s, genes encoding Bt insecticidal proteins were expressed in crop plants such as cotton and corn to protect these crops from insect damage. The first Bt protein used in transgenic cotton was Cry1Ac to control Heliothis virescens (tobacco budworm). Cry1Ab was applied to corn to control Ostrinia nubilalis (European corn borer). Since these insects have developed resistance to Cry1Ac and Cry1Ab, new Bt proteins are required to overcome the resistance. In order to protect corn furthermore, it is desired to control Diabrotica virgifera (Western corn rootworm), Helicoverpa zea (corn earworm) and Spodoptera frugiperda (fall armyworm). Recently, many new Bt insecticidal proteins have been discovered, but most of them require protein engineering to meet the high activity standard for commercialization. The engineering process for higher activity necessary for Bt crops is called optimization. The seed industry has been optimizing Bt insecticidal proteins to improve their insecticidal activity. In this review, several optimization projects, which have led to substantial activity increases of Bt insecticidal proteins, are described.
