Chemical agents have been used to control JPS beetle, but due to various issues, efforts are being made to replace these chemical agents with entomopathogenic fungi. We investigated the expression of immune-related genes in JPS beetle in response to infection with JEF-197, a Metarhizium anisopliae isolate, using RNA-seq. RNA samples were obtained from JEF-197, JPS adults treated with JEF-197, and non-treated JPS adults on the 8th day after fungal treatment, and RNA-seq was performed using Illumina sequencing. JPS beetle transcriptome was assembled de novo and differentially expressed gene (DEG) analysis was performed. There were 719 and 1953 up- and downregulated unigenes upon JEF-197 infection, respectively. Upregulated contigs included genes involved in RNA transport, ribosome biogenesis in eukaryotes, spliceosome-related genes, and genes involved in immune-related signaling pathways such as the Toll and Imd pathways. Forty-two fungal DEGs related to energy and protein metabolism were upregulated, and genes involved in the stress response were also upregulated in the infected JPS beetles. Together, our results indicate that infection of JPS beetles by JEF-197 induces the expression of immune-related genes. The validation of a previously developed model of the interaction between the red pigment-concentrating hormone of Daphnia pulex and its cognate receptor (Jackson et al., IJBM 106, 969-978, 2018) was undertaken. Single amino acid replacements, noticeably an Ala scan, of the ligand, Dappu-RPCH, were docked to the receptor, and the binding energies calculated and compared to the one with Dappu-RPCH. As a second step, the same molecules were docked using molecular dynamics (MD) in a 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) membrane. Changes in binding energy were compared to previous results on in vitro receptor activation (Marco et al., Sci. Rep. 7, 6851, 2017). Residue scanning and MD simulations both gave comparable results for binding energy. For most mutants, there was a good inverse correlation between in vitro activity and binding. There were, however, exceptions; for example [Ala4]Dappu-RPCH bound as tightly as the cognate ligand but had little activity. This seeming discrepancy was explained when the MD data were analyzed in detail, showing that, although [Ala4]Dappu-RPCH had multiple interactions with the receptor accounting for the high binding energy, the interacting residues of the receptor were quite different to those of Dappu-RPCH. The MD calculations show clearly that the strong binding affinity of the ligand to the receptor is not sufficient for activation. Interaction of the binding of the ligand to two residues of the receptor, Ser 155 and Gln 237, is also essential. A comparison of our computational results with the experimental results of Marco et al. and comparison with the extensive data on GnRH supports the validity of our Dappu-RPCH R model.White matter atrophy has been shown to precede the massive loss of striatal GABAergic neurons in Huntington's disease (HD). This study investigated the effects of in vivo expression of reprogramming factor octamer-binding transcription factor 4 (OCT4) on neural stem cell (NSC) niche activation in the subventricular zone (SVZ) and induction of cell fate specific to the microenvironment of HD. R6/2 mice randomly received adeno-associated virus 9 (AAV9)-OCT4, AAV9-Null, or phosphate-buffered saline into both lateral ventricles at 4 weeks of age. The AAV9-OCT4 group displayed significantly improved behavioral performance compared to the control groups. Following AAV9-OCT4 treatment, the number of newly generated NSCs and oligodendrocyte progenitor cells (OPCs) significantly increased in the SVZ, and the expression of OPC-related genes and glial cell-derived neurotrophic factor (GDNF) significantly increased. Further, amelioration of myelination deficits in the corpus callosum was observed through electron microscopy and magnetic resonance imaging, and striatal DARPP32+ GABAergic neurons significantly increased in the AAV9-OCT4 group. These results suggest that in situ expression of the reprogramming factor OCT4 in the SVZ induces OPC proliferation, thereby attenuating myelination deficits. Particularly, GDNF released by OPCs seems to induce striatal neuroprotection in HD, which explains the behavioral improvement in R6/2 mice overexpressing OCT4.Bio-based monomers represent the future market for polymer chemistry, since the political economics of different states promote green ventures toward more sustainable materials and processes. Industrial pulp and paper processing represent a large market that could advance the use of by-products to avoid waste production and reduce pollution. Lignin represents the most available side product that can be used to produce a bio-based monomer. This review is concentrated on the possibility of using bio-based monomer derivates from pulp and the paper industry for UV-curing processing. UV-curing represents the new frontier for thermoset production, allowing a fast reaction cure, less energy demand, and the elimination of solvent. The growing demand for new monomers increases research in the environmental field to substitute for petroleum-based products. This review provides an overview of the main monomers and relative families of compounds derived from industrial processes that are suitable for UV-curing. Particular focus is given to the developments reached in the last few years concerning lignin, rosin and terpenes and the related possible applications of these in UV-curing chemistry. In head and neck cancer management, there is a need for tailored approaches to optimally implement clinical outcomes. Based on the assumption that efficacy and long-term toxicity are not satisfactory for standard concurrent platinum-based chemoradiotherapy, several trials have been designed to test whether induction immunotherapy and/or concomitant immunotherapy and radiotherapy result in improved survival and toxicity outcomes. Here, we present an overview of the most recent concomitant therapeutic strategies for head and neck cancer, focusing on the knowledge available regarding check-point inhibitors. The aim is to present the characteristics of the main check-point inhibitors and to summarize the clinical trials on the combination of immune check-point inhibitors and (chemo)radiotherapy in the definitive HNC setting, in order to provide a useful clinical tool for further research.Since late 2019, Coronavirus Disease 2019 (COVID-19) has spread all over the world. The disease is highly contagious, and it may lead to acute respiratory distress (ARD). Medical imaging can play an important role in classifying, detecting, and measuring the severity of the virus. This study aims to provide a novel auto-detection tool that can detect abnormal changes in conventional X-ray images for confirmed COVID-19 cases. X-ray images from patients diagnosed with COVID-19 were converted into 19 different colored layers. Each layer represented objects with similar contrast that could be defined as a specific color. The objects with similar contrasts were formed in a single layer. All the objects from all the layers were extracted as a single-color image. Based on the differentiation of colors, the prototype model was able to recognize a wide spectrum of abnormal changes in the image texture. This was true even if there was minimal variation of the contrast values of the detected uncleared abnormalities. The results indicate that the proposed novel method can detect and determine the degree of lung infection from COVID-19 with an accuracy of 91%, compared to the opinions of three experienced radiologists. The method can also efficiently determine the sites of infection and the severity of the disease by classifying the X-rays into five levels of severity. Thus, the proposed COVID-19 autodetection method can identify locations and indicate the degree of severity of the disease by comparing affected tissue with healthy tissue, and it can predict where the disease may spread. This paper reports on a new approach to face verification in long-wavelength infrared radiation. Two face images were combined into one double image, which was then used as an input for a classification based on neural networks. For testing, we exploited two external and one homemade thermal face databases acquired in various variants. The method is reported to achieve a true acceptance rate of about 83%. We proved that the proposed method outperforms other studied baseline methods by about 20 percentage points. We also analyzed the issue of extending the performance of algorithms. We believe that the proposed double image method can also be applied to other spectral ranges and modalities different than the face. The application of natural fibers is rapidly growing in many sectors, such as construction, automobile, and furniture. Kenaf fiber (KF) is a natural fiber that is in demand owing to its eco-friendly and renewable nature. Nowadays, there are various new applications for kenaf, such as in absorbents and building materials. It also has commercial applications, such as in the automotive industry. Magnesium hydroxide (Mg(OH)2) is used as a fire retardant as it is low in cost and has good flame retardancy, while polyester yarn (PET) has high tensile strength. The aim of this study was to determine the horizontal burning rate, tensile strength, and surface morphology of kenaf fiber/PET yarn reinforced epoxy fire retardant composites. The composites were prepared by hybridized epoxy and Mg(OH)2 PET with different amounts of KF content (0%, 20%, 35%, and 50%) using the cold press method. The specimen with 35% KF (epoxy/PET/KF-35) displayed better flammability properties and had the lowest average burning rate of 14.55 mm/min, while epoxy/PET/KF-50 with 50% KF had the highest tensile strength of all the samples. This was due to fewer defects being detected on the surface morphology of epoxy/PET/KF-35 compared to the other samples, which influenced the mechanical properties of the composites. The therapeutic use of peptides has increasingly recognized in the development of new therapies. read more However, the susceptible enzymatic cleavage is a barrier that needs to overcome. Nose-to-brain delivery associated with liposomes can protect peptides against biodegradation and improve the accessibility to brain targets. The aim was to develop a liposomal formulation as ghrelin carrier. The quality by design (QbD) approach was used as a strategy for method development. The initial risk assessments were carried out using a fishbone diagram. A screening design study was performed for the critical material attributes/critical process parameters (CMAs/CPPs) on critical quality attributes (CQAs). Liposomes were obtained by hydrating phospholipid films, followed by extrusion or homogenization, and coated with chitosan. The optimized liposome formulation was produced by high-pressure homogenization coated with chitosan, and the resulted were liposomes size 72.25 ± 1.46 nm, PDI of 0.300 ± 0.027, the zeta potential of 50.
