Background Oral squamous cell carcinoma (OSCC) is the most common malignant tumor of the oral cavity. The abundance of cell proliferation marker Ki-67, epithelial-mesenchymal transition (EMT) markers E-cadherin, N-cadherin, and vimentin was analyzed by western blot assay. Glycolysis was evaluated using commercial kits. The interaction between hsa_circ_0072387 and miR-503-5p was confirmed by bioinformatics analysis, RNA immunoprecipitation (RIP) assay, and dual-luciferase reporter assay. Results hsa_circ_0072387 expression was significantly downregulated, and miR-503-5p was upregulated in OSCC cells and tissues. Gain of hsa_circ_0072387 or knockdown of miR-503-5p suppressed the cell proliferation, migration and invasion, EMT, and glycolysis in OSCC SCC-4 and HSC-3 cells. hsa_circ_0072387 targeted miR-503-5p and inversely regulated miR-503-5p expression. Moreover, upregulation of miR-503-5p could partially revert the tumor-suppressive effects of hsa_circ_0072387 on OSCC cells. Conclusion hsa_circ_0072387 inhibited OSCC progression by downregulating miR-503-5p, explicating that hsa_circ_0072387 could function as a novel potential therapeutic target for OSCC.Aliphatic di- and polyisocyanates are crucial chemical ingredients in many industrial processes and are a well-recognized cause of occupational asthma. Serologic detection of "chemical epitopes" in biological samples could serve as an exposure surveillance approach toward disease prevention, and thus we sought to generate aliphatic isocyanate-specific monoclonal antibodies (mAbs). Three hybridomas were generated from Balb/c mice immunized with a commercial product containing a combination of uretdione, homopolymer, and monomeric forms of hexamethylene diisocyanate (HDI). Three stable hybridomas were subcloned by limiting dilution, two secreting IgG1κ and one secreting IgMκ mAb that bind aliphatic di- and polyisocyanates (conjugated to albumin), but not aromatic toluene or methylene diphenyl diisocyanate (TDI or MDI). Each mAb demonstrates slight differences in epitope specificity, for example, recognition of hydrogenated MDI (HMDI) or different carrier proteins (transferrin, actin) reacted with vapor phase HDI, and is encoded by unique recombination of different germline antibody genes, with distinct complementary determining regions. By western blot, all three mAbs detect a molecule with characteristics of an albumin adduct uniquely in urine from mice skin exposed to a mixture of aliphatic di- and polyisocyanate. Together, the data define molecular determinants of humoral immune recognition of aliphatic di- and polyisocyanates through new mAbs, which will serve as useful research reagents and may be applicable to future exposure surveillance efforts.Background Medical assistance in dying (MAID) became legal in Canada in 2016. Although the legislation is federal, each province is responsible for establishing quality care. Objective To explore patient, family, and health care provider (HCP) perspectives on MAID access and care delivery and improve regional MAID care delivery. Design Qualitative exploratory. Setting/Subjects We interviewed 5 patients (4 met the legislated MAID criteria and 1 did not), 11 family members (4 spouses, 5 children, 1 sibling, and 1 friend), and 14 HCP (3 physicians, 4 social workers, and 7 nurses) from June to August 2017. Measurement Semistructured interviews, content analysis, and thematic summary. Results Patients, families, and HCPs highlighted access and delivery concerns regarding program sustainability, care pathway ambiguity, lack of support for care choices, institutional conscientious objection (CO), navigating care in institutions with a CO, and postdeath documentation. Patients and families expressed additional concerns regarding lack of ability to provide advanced MAID consent, and the requirement of independent witnesses on MAID request forms and consent immediately before MAID administration. Conclusion Quality improvement (QI) processes are essential to devise an accessible dignified patient- and family-centered MAID program. Ensuring patient and family perspectives are integrated into QI initiatives will assist programs in ensuring the needs of all are considered in structuring and staffing a program that is accessible, easy to navigate, and provides dignified end-of-life care in supportive and respectful work environments.Vitamin B12 plays an important role in cell division and is of vital importance during pregnancy. Iron and B12 deficiency increase the risk of neonatal morbidity and the outcome of the overall pregnancy. The aim of our study was to analyze whether the use of vitamin B12, with standard supplements of folic acid and iron among nonanemic pregnant women, will result in improvements of hemogram parameters in terms of hematological and biochemical markers. Study participants were 200 healthy pregnant women, randomized into an intervention group and a control group, recruited from gynecological primary care practices in Split, Croatia. In addition to standard supplementation (350 mg/day ferrous iron, 5 mg folic acid), participants in the intervention group were given 5 μg of vitamin B12 each morning for 100 days. Both biochemical and hematological measurings were conducted in two intervals 8th-10th week of gestation and then again in the 34th-36th week of gestation. Participants in the control group were given only standard-of-care iron and folic acid supplementation.
