The repurposing of approved drugs presents a promising strategy to address current bottlenecks in the development of novel antibacterial agents. Drug repurposing is a cost-effective emerging strategy, which aims to treat resistant infectious diseases by identifying known drugs with predicted efficacy for diseases other than the target disease. This strategy has potential in the treatment of tuberculosis (TB), particularly drug-resistant TB. In recent years, a panel of drugs approved for clinical use or clinical trials, such as linezolid, vancomycin and celecoxib, have been found to have anti-TB activities. However, the utility of drug repurposing is limited by the number of candidate compounds and their low activities. The low activities of repurposed drugs have slowed the development of a drug-repurposing strategy for anti-TB drugs. The present review discusses progress in the discovery of new anti-TB agents through drug repurposing since 2014. We also discuss the challenges faced and analyze the innovative ways that are being used to overcome these difficulties. This review may provide a useful guide for researchers in the field of drug repurposing. Buruli ulcer is an emerging tissue-necrosis infectious disease, caused by the pathogen Mycobacterium ulcerans, leading to permanent deformity if untreated. Despite this debilitating condition, no specific disease-modifying therapeutics or vaccination is available to date. Therefore, we aimed to design an effective multi-epitope vaccine against M. ulcerans using vaccinomics approach. Briefly, the highest antigenic PE-PGRS protein was selected from which the promiscuous T- and B-cell epitopes were predicted. After rigorous assessment, 15 promising T- and B-cell epitopes were selected. The identified T-cell epitopes showed marked interactions towards their HLA-binding alleles and provided 99.8 % world population coverage. Consequently, a vaccine chimera was designed by connecting these epitopes with suitable linkers and LprG adjuvant. The vaccine construct was highly antigenic, immunogenic and non-allergenic; hence, subjected to homology modelling. The molecular docking and dynamics simulation revealed a strong and stable interaction between vaccine and toll-like receptor 2. The binding energy and dissociation constant were -15.3 kcal/mol and 5.9 × 10-12 M, respectively. The computer-simulated immune responses showed abundance of immunoglobulins, increased interferon-γ production, and macrophages activation which are crucial for immune response against M. ulcerans. Furthermore, disulfide bridging and in silico cloning were also performed. These results suggest that the vaccine, if validated experimentally, will be a promising candidate against M. ulcerans and prevent Buruli ulcer disease. BACKGROUND AND OBJECTIVE Conventional anthropometric studies using Kinect depth sensors have concentrated on estimating the distances between two points such as height. This paper deals with a novel waist measurement method using SVM regression, further widening spectrum of Kinect's potential applications. Waist circumference is a key index for the diagnosis of abdominal obesity, which has been linked to metabolic syndromes and other related diseases. Yet, the existing measuring method, tape measure, requires a trained personnel and is therefore costly and time-consuming. METHODS A dataset was constructed by recording both 30 frames of Kinect depth image and careful tape measurement of 19 volunteers by a clinical investigator. This paper proposes a new SVM regressor-based approach for estimating waist circumference. A waist curve vector is extracted from a raw depth image using joint information provided by Kinect SDK. To avoid overfitting, a data augmentation technique is devised. The 30 frontal vectors and proach of delivering care without laying burdens on patients. OBJECTIVES Spring ligament (SL) insufficiency is observed commonly in association with adult-acquired flatfoot deformity. Evaluation of the SL using ultrasonography (US) has not been fully investigated, and the functional characteristics of the spring ligament have not been clarified. The purpose of this study was to perform a functional evaluation of the SL using US. METHODS Fifty-one healthy young volunteers (31 males, 20 females; 102 feet) participated in this study. The thickness of the SL in both non-weight bearing (NSL) and 90% weight bearing (WSL) conditions was measured using US. The correlation between assessment of foot alignment (Navicular Drop test (NDT), Arch height Index (AHI), and Arch height flexibility (AHF)) and the thickness of the SL was investigated. RESULTS The thickness of the SL in NSL and WSL conditions was 2.28 ± 0.38 mm and 2.13 ± 0.34 mm, respectively. The thickness was 2.42 ± 0.38 mm (NSL)/2.26 ± 0.31 mm (WSL) in males, and 2.07 ± 0.28 mm (NSL)/1.93 ± 0.29 mm (WSL) in females. The standard values of the thickness of the SL were 2.0-2.8 mm in males and 1.8-2.4 mm in females. The thickness of the SL was significantly different between males and females (p less then 0.01), but was within the margin of error between NSL and WSL. The relationship between NSL and foot alignment only showed a weak correlation with AHI (r = 0.23, p less then 0.05). CONCLUSIONS Our results indicate that the SL is a hardy structure that shows little change in thickness on weight bearing in vivo. Black rice is rich in phenolic acids and anthocyanin; however, limited studies have determined its effect on ground beef quality. The objective was to determine the effects of 0, 0.4, 0.8, and 1.2% black rice water extract (BRWE) on ground beef patties quality when packaged in polyvinyl chloride (PVC). pH, surface color, lipid oxidation, total plate count, and antioxidant capacity were determined on 0, 3, and 6 days of storage under fluorescent light at 2 °C. Addition of BRWE had no effect (P = .98) on pH. Incorporating BRWE in ground beef improved (P less then .0001) redness compared with control. Eganelisib manufacturer The addition of BRWE decreased (P less then .0001) lipid oxidation compared with control during storage; while antioxidant capacity increased with the addition of extract. BRWE at 1.2% reduced (P = .007) aerobic microbial counts after 6 days of storage. These results suggested that BRWE could be used as a natural antioxidant in ground beef to limit lipid oxidation and discoloration.
