This study was performed to investigate role and mechanism of lncRNA Rian (RNA imprinted and accumulated in nucleus) in sevoflurane anesthesia-induced cognitive dysfunction. However, intracerebroventricularly injection with Adenovirus (Ad) for the over-expression of Rian ameliorated sevoflurane-induced neuronal damage and apoptosis. Cognitive impairments induced by sevoflurane were attenuated by injection with Ad-Rian. Moreover, transfection with Ad-Rian also protected isolated primary hippocampal neurons against sevoflurane-induced decrease of cell viability and increase of lactic acid dehydrogenase (LDH) and apoptosis. Mechanistically, Rian bind to miR-143-3p, and decreased expression of LIMK1 (Lim kinase 1) through negative regulation of miR-143-3p. Knockdown of LIMK1 aggravated sevoflurane-induced decrease of cell viability and increase of LDH and apoptosis in neurons, while over-expression attenuated LIMK1 silence-induced neuronal damage post-sevoflurane anesthesia. In conclusion, Rian demonstrated neuroprotective effects against sevoflurane anesthesia-induced cognitive dysfunction through regulation of miR-143-3p/LIMK1 axis, providing promising target for sevoflurane anesthesia-induced cognitive dysfunction.The production of red blood cells in vitro, which is useful for basic or clinical research, has been improved. link2 Further optimization of culture protocols may facilitate erythroid differentiation from hematopoietic stem cells to red blood cells. However, the details of erythropoiesis, particularly regarding the behaviors of differentiation-related proteins, remain unclear. Here, we performed erythroid differentiation using two independent bone marrow- or cord blood-derived CD34+ cell sources and identified proteins showing reproducible differential expression in all groups. Notably, most of the proteins expressed at the early stage were downregulated during erythroid differentiation. However, seven proteins showed upregulated expression in both bone marrow cells and cord blood cells. These proteins included alpha-synuclein and selenium-binding protein 1, the roles of which have not been clarified in erythropoiesis. There is a possibility that these factors contribute to erythroid differentiation as they maintained a high expression level. Retro-cues, displayed after the offset of a memory stimulus and prior to the onset of a probe stimulus, indicate the test item and improve performance in VSTM tasks. It has been proposed that retro-cues aid recall by transferring information from a high-capacity memory store into visual working memory (multiple-store hypothesis). Alternatively, retro-cues could aid recall by redistributing memory resources within the same (low-capacity) working memory store (single-store hypothesis). If retro-cues provide access to a memory store with a capacity exceeding the set size, then, given sufficient training in the use of the retro-cue, near-ceiling performance should be observed. To test this prediction, 10 observers each performed 12 hours across 8 sessions in a retro-cue change-detection task (40,000+ trials total). The results provided clear support for the single-store hypothesis retro-cue benefits (difference between a condition with and without retro-cues) emerged after a few hundred trials and then remained constant throughout the testing sessions, consistently improving performance by two items, rather than reaching ceiling performance. Surprisingly, we also observed a general increase in performance throughout the experiment in conditions with and without retro-cues, calling into question the generalizability of change-detection tasks in assessing working memory capacity as a stable trait of an observer (data and materials are available at osf.io/9xr82 and github.com/paulzerr/retrocues). In summary, the present findings suggest that retro-cues increase capacity estimates by redistributing memory resources across memoranda within a low-capacity working memory store.Although visual and auditory inputs are initially processed in separate perception systems, studies have built on the idea that to maintain spatial information these modalities share a component of working memory. The present study used working memory navigation tasks to examine functional similarities and dissimilarities in the performance of updating tasks. Participants mentally updated the spatial location of a target in a virtual array in response to sequential pictorial and sonant directional cues before identifying the target's final location. We predicted that if working memory representations are modality-specific, mixed-modality cues would demonstrate a cost of modality switching relative to unimodal cues. The results indicate that updating performance using visual unimodal cues positively correlated with that using auditory unimodal cues. Task performance using unimodal cues was comparable to that using mixed modality cues. link3 The results of a subsequent experiment involving updating of target traces were consistent with those of the preceding experiments and support the view of modality-nonspecific memory.
Patient-reported outcome measures (PROMs) are used to capture patient perspectives in disease assessment. The objective of this study was to capture feedback about commonly used PROMs for spondyloarthritis (SpA) through semi-structured group discussions with individuals diagnosed with psoriatic arthritis (PsA) or ankylosing spondylitis (AS). The goal was to identify PROM content that most resonated with patient experiences and is therefore suitable for implementation in SpA clinical practice.

Semi-structured tasks and probes were designed to elicit qualitative patient feedback on several general health and disease-specific PROMs. During a series of in-person and telephone meetings, participants with PsA or AS were asked to identify content that resonated with them and to identify items that may not have captured their personal experiences living with their disease. Both individualized and small group review and concept elicitation were captured after participant review of PROMs.

Both PsA and AS participcision-making conversations between patients and their healthcare providers. Participants indicated that constructs such as isolation, depression, fatigue, and relationships with others were critical to inform healthcare professionals about the patient experience of living with their disease.

Eribulin mesylate (eribulin) is an efficient microtubule inhibitor that is used for metastatic breast cancer. However, breast cancer can develop resistance to eribulin. This resistance mechanism needs to be elucidated.

A transposon mutagenesis screen was conducted using a pPB-SB-CMV-puro-SD plasmid and pCMV-PBase transposase. Viability and cytotoxicity were analyzed by MTT assay and flow cytometry, respectively. Real-time PCR and western blot were used for gene expression analysis. In addition, vivo study was also designed to analyze therapy efficiency.

TAB2, which is part of the nuclear factor-kappa B (NF-κB) pathway, was identified as a candidate eribulin-resistant gene. TAB2 down-regulation resulted in significantly lower cell viability and higher cytotoxicity of cells treated with eribulin, while TAB2 up-regulation showed opposite results. Similarly, combination of NF-κB inhibitors [Bay-117082 and QNZ (quinazoline derivative)] with eribulin showed significantly lower cell viability and higher drug cytotoxicity than single agent treatment with eribulin in MDA-MB-231 cells.
