This study investigated anaerobic co-digestion in semi-continuous reactors of biowastes (75 % Volatil Solids) and biodegradable plastics (25 % Volatil Solids, PLA and PHB). PHB was estimated to be fully biodegraded in the reactors. By contrast, PLA accumulated in the reactor, and an average biodegradation of 47.6 ± 17.9 % was estimated during the third hydraulic retention time. Pretreatment of PLA, by thermo-alkaline hydrolysis at 70 °C, with 2.5 w/v of Ca(OH)2 for 48 h, improved the biodegradation yield of PLA to 77.5 ± 9.3 %. Finally, it was highlighted that PLA or PHB addition to the feed did not further affect the agronomic properties of the digestate.This study investigated the reduction of nitrate in a mesophilic waste activated sludge (WAS) fermentation system and determined the effect of nitrate reduction on the hydrolysis, acidogenesis and acetogenesis. Experimental results showed that the initial nitrate concentrations of 100, 200 and 400 mg/L were completely reduced in 1, 2 and 7 days, respectively. The destruction of volatile suspended solids was 1.2, 1.8 and 2.8 times, respectively, that without nitrate, demonstrating nitrate promoted the release of organic matter in sludge and enhanced the biodegradability of sludge organics. Moreover, batch tests using model substrates illustrated nitrate reduction promoted sludge hydrolysis and acetogenesis, but slightly inhibited acidogenesis. This study offers a feasible method to address two major problems currently faced by biological wastewater treatment plants, i.e. the overabundance of WAS and the lack of carbon sources for the denitrification process.Reduction of inherent structural recalcitrance and improved saccharification efficiency are two important facets to enhance fermentable sugar yield for bioethanol production from lignocellulosic biomass. This study optimized alkaline pretreatment and saccharification conditions employing response surface methodology to improve saccharification yield of jute (Corchorus olitorius cv. JROB-2) biomass. The biomass is composed of cellulose (66.6 %), lignin (19.4 %) and hemicellulose (13.1 %). NaOH concentration exhibited significant effect on delignification during pretreatment. The highest delignification (80.42 %) was obtained by pretreatment with 2.47 % NaOH at 55.8 °C for 5.9 h removing 79.8 % lignin and 34.2 % hemicellulose from biomass, thereby increasing cell wall porosity and allowing better accessibility to saccharification enzyme. During saccharification optimization, significant effect was observed for biomass loading, enzyme concentration and temperature. Optimized saccharification condition yielded maximum saccharification (76.48 %) when hydrolysis was performed at 6.9 % biomass loading with enzyme concentration of 49.52 FPU/g substrate at 51.05 °C for 74.46 h.The organic fraction of municipal solid waste (OFMSW) is a widely-available promising feedstock for biofuel production. However, the presence of different inhibitors originating from fruit and food/beverage wastes as well as recalcitrant lignocellulosic fractions hampers its bioconversion. This necessitates a pretreatment to augment the biodigestibility and fermentability of OFMSW. Hence, this review aims to provide the in-vogue inhibitory compound removal and pretreatment techniques that have been employed for efficient OFMSW conversion into biofuels, i.e., hydrogen, biogas, ethanol, and butanol. The techniques are compared concerning their mode of action, chemical and energy consumption, inhibitor formation and removal, economic feasibility, and environmental sustainability. This critique also reviews the existing knowledge gap and future perspectives for efficient OFMSW valorization. The insights provided pave the way toward developing energy-resilient cities while addressing environmental crises related to generating OFMSW.Effective inducers play essential roles in the regulation of cell growth and astaxanthin production in Haematococcus pluvialis. Here, a novel 5-aminolevulinic acid (5-ALA) feeding strategy was developed and found to enhance H. pluvialis biomass, fatty acid, and astaxanthin yields. Specifically, 5-ALA feeding (4 μM) on day 1 caused respective 23.8 %, 24.8 %, and 20.3 % increases in biomass, fatty acid, and astaxanthin yields. The observed enhancement of biomass accumulation associated with the provision of 5-ALA during the early stages of growth was attributable to enhanced photosynthetic carbon assimilation. This increased biomass accumulation, in turn, contributed to the measured increases in both fatty acid and astaxanthin yields. Overall, these results provide new insight into the importance of photosynthetic carbon assimilation as a determinant of biomass accumulation and a regulator of both fatty acid and astaxanthin production, while offering an effective strategy capable of accelerating astaxanthin production by H. pluvialis in commercial agricultural contexts.Paddy straw (PS) and pine needles (PN) are one of the challenging biomasses in terms of disposal and compost making due to their high silica and tannin contents. Particulate air pollution, loss of biodiversity and respiratory impairments are some of disastrous outcomes caused by burning. However, high percentage of cellulose and hemicellulose makes them potential substrate for paper and pulp industries. The main aim of work was to study and utilize a combinatorial approach of weak chemical treatment and lignin degrading fungal species as agents of effective production of lignin modifying enzymes (LME's) for lignin depolymerisation from the biomasses. Phanerochaete chrysosporium was found to be the best degrader of lignin (47.11 % in PS + PN in 28 days) with maximum LME's production between 10th-17th days. Efficient lignin degradation in the PS and PN biomass will aid further application in pulp production supporting the transition to a circular economy in a greener way.Despite the development and incorporation of new therapeutic strategies, such as biologic therapy and small molecules, corticosteroids still play an important role in inducting inflammatory bowel diseases (IBD) remission. Variables like indicating the right doses at the right time, in adequate intervals, the security of these drugs and the pharmacological alternatives available must be considered by the providers when they are indicated to patients with IBD. Although the use of corticosteroids is considered as a marker of quality of care in patients with IBD, the use of these drugs in the clinical practice of IBD is far from being the correct one. This review article is not intended to be just a classic review of the indications for corticosteroids. Here we explain the scenarios in which, in our opinion, steroids would not be an appropriate option for our patients, as well as the most frequent mistakes we make in our daily practice when using them.
Loss-of-response and adverse events (AE) to biologics have been linked to HLA-DQA1*05 allele. However, the clinical factors or biologic used may influence treatment duration. Our objective was to evaluate the influence of clinical and therapeutic factors, along with HLA, in biological treatment discontinuation.

A retrospective study of consecutive IBD patients treated with biologics between 2007 and 2011 was performed. Main outcome was treatment discontinuation due to primary non-response (PNR), secondary loss of response (SLR) or AE. HLA-DQA1 genotyping was done in all patients. Regression analyses were used to assess risk factors of treatment discontinuation.

One hundred fifty patients (61% male) with 312 biologic treatments were included. 147 (47%) were discontinued with a cumulative probability of 30%, 41% and 56% at 1, 2 and 5 years. In the present essay we first review the extensive evidence that indicates this modulation is mediated by adenosine A1 and A2A receptors (A1Rs and A2ARs) differentially expressed by the components of the striatal microcircuit that include cortico-striatal glutamatergic and mesencephalic dopaminergic terminals, and the cholinergic interneuron. This microcircuit mediates the ability of striatal glutamate release to locally promote dopamine release through the intermediate activation of cholinergic interneurons. Daclatasvir in vivo A1Rs and A2ARs are colocalized in the cortico-striatal glutamatergic terminals, where they form A1R-A2AR and A2AR-cannabinoid CB1 receptor (CB1R) heteromers. We then evaluate recent findings on the unique properties of A1R-A2AR and A2AR-CB1R heteromers, which depend on their different quaternary tetrameric structure. These properties involve different allosteric mechanisms in the two receptor heteromers that provide fine-tune modulation of adenosine and endocannabinoid-mediated striatal glutamate release. Finally, we evaluate the evidence supporting the use of different heteromers containing striatal adenosine receptors as targets for drug development for neuropsychiatric disorders, such as Parkinson's disease and restless legs syndrome, based on the ability or inability of the A2AR to demonstrate constitutive activity in the different heteromers, and the ability of some A2AR ligands to act preferentially as neutral antagonists or inverse agonists, or to have preferential affinity for a specific A2AR heteromer.Neuroinflammation, specifically the NLRP3 inflammasome cascade, is a common underlying pathological feature of many neurodegenerative diseases. C101248 showed a concentration-dependent inhibition of both mouse and human THIK-1 (IC50 ∼50 nM) and was inactive against K2P family members TREK-1 and TWIK-2, and Kv2.
