These newly defined alterations may provide possibilities for innovative therapeutic approaches.In French Guiana, canine heartworm disease is well known, but the diversity of filarial parasites of dogs remains largely unknown. A total of 98 canine blood samples from Cayenne and Kourou were assessed by a blood wet mount preparation, heartworm antigen test and molecular exploration of filarioid and Wolbachia DNAs, followed by a multiplex species-specific qPCR's identification and a subsequent sequencing analysis. Thereafter, a phylogeny based on maximum likelihood was carried out to facilitate specific identification. Five dogs were microfilaremic. Heartworm antigens were detected in 15 (15.3%) dogs. Of these, six (6.1%) were considered as occult infections as neither microfilariae nor Dirofilaria immitis DNA were detected. The 11 (11.2%) D. immitis isolates corresponded to a low virulent strain. Six of the D. immitis isolates were positive for Wolbachia endosymbionts of D. immitis belonging to the clade C DNA. Acanthocheilonema reconditum DNA was detected in 3 (3.1%) samples. Of these latter, one was found co-infected with the Brugia sp. genotype and the DNA of the clade D of the Wolbachia endosymbiont of Brugia species. This latter was also detected in two filarioid DNA-free samples. Finally, two samples were positive for Cercopithifilaria bainae genotype, which is distinct from those identified in Europe. The present study highlights the urgent need to implement chemoprophylaxis associated with anti-Wolbachia drugs to control these potential zoonoses.Melanoma is the most aggressive skin cancer with an extremely challenging therapy. The dermal-epidermal junction (DEJ) degradation and subsequent dermal invasion are the earliest steps of melanoma dissemination, but the mechanisms remain elusive. We previously identified Tspan8 as a key actor in melanoma invasiveness. Here, we investigated Tspan8 mechanisms of action during dermal invasion, using a validated skin-reconstruct-model that recapitulates melanoma dermal penetration through an authentic DEJ. We demonstrate that Tspan8 is sufficient to induce melanoma cells' translocation to the dermis. Mechanistically, Tspan8+ melanoma cells cooperate with surrounding keratinocytes within the epidermis to promote keratinocyte-originated proMMP-9 activation process, collagen IV degradation and dermal colonization. This concurs with elevated active MMP-3 and low TIMP-1 levels, known to promote MMP-9 activity. The study includes 32 horses, divided into three groups "young" adult group (aged 8-16 years old) "geriatric" group (aged 18-24 years old) and the "PPID" group (aged 15-31 years old). The PPID group was further divided into two subgroups PPID ID+ and PPID ID- based on presence or absence of ID. We measured serum antioxidant stress markers in all horses total oxidant status (TOS), total antioxidant capacity (TAC), ceruloplasmin (CER), lipofuscin (LPS), malondialdehyde (MDA) and thiols concentrations (containing sulfhydryl group -SH) as well as enzymatic systems total superoxide dismutase (SOD), cytoplasmic SOD (CuZnSOD), mitochondrial SOD activity (MnSOD). Total serum thiols were significantly lower in the geriatric group and in the PPID group compared to the young group. The MnSOD concentration was higher in the PPID ID+ group compared to the PPID ID-. LPS and MDA concentrations were lower in the PPID ID+ group compared to the PPID ID- group. In the selected study groups of horses, older age, the presence of PPID and ID in the case of PPID had no effect on the studied oxidative stress markers.Investigating the patterns of homozygosity, linkage disequilibrium, effective population size and inbreeding coefficients in livestock contributes to our understanding of the genetic diversity and evolutionary history. Here we used Illumina PorcineSNP50 Bead Chip to identify the runs of homozygosity (ROH) and estimate the linkage disequilibrium (LD) across the whole genome, and then predict the effective population size. In addition, we calculated the inbreeding coefficients based on ROH in 305 Piétrain pigs and compared its effect with the other two types of inbreeding coefficients obtained by different calculation methods. A total of 23,434 ROHs were detected, and the average length of ROH per individual was about 507.27 Mb. There was no regularity on how those runs of homozygosity distributed in genome. The comparisons of different categories suggested that the formation of long ROH was probably related with recent inbreeding events. Although the density of genes located in ROH core regions is lower than that in the other genomic regions, most of them are related with Piétrain commercial traits like meat qualities. Overall, the results provide insight into the way in which ROH is produced and the identified ROH core regions can be used to map the genes associated with commercial traits in domestic animals.Chitosan is increasingly used for safe nucleic acid delivery in gene therapy studies, due to well-known properties such as bioadhesion, low toxicity, biodegradability and biocompatibility. Furthermore, chitosan derivatization can be easily performed to improve the solubility and stability of chitosan-nucleic acid polyplexes, and enhance efficient target cell drug delivery, cell uptake, intracellular endosomal escape, unpacking and nuclear import of expression plasmids. As in other fields, chitosan is a promising drug delivery vector with great potential for the fish farming industry. This review highlights state-of-the-art assays using chitosan-based methodologies for delivering nucleic acids into cells, and focuses attention on recent advances in chitosan-mediated gene delivery for fish biotechnology applications. The efficiency of chitosan for gene therapy studies in fish biotechnology is discussed in fields such as fish vaccination against bacterial and viral infection, control of gonadal development and gene overexpression and silencing for overcoming metabolic limitations, such as dependence on protein-rich diets and the low glucose tolerance of farmed fish. Finally, challenges and perspectives on the future developments of chitosan-based gene delivery in fish are also discussed.In various vascular diseases, extracellular matrix (ECM) and integrin expression are frequently altered, leading to focal adhesion kinase (FAK) or proline-rich tyrosine kinase 2 (Pyk2) activation. In addition to the major roles of FAK and Pyk2 in regulating adhesion dynamics via integrins, recent studies have shown a new role for nuclear FAK in gene regulation in various vascular cells. In particular, FAK primarily localizes within the nuclei of vascular smooth muscle cells (VSMCs) of healthy arteries. However, vessel injury increased FAK localization back to adhesions and elevated FAK activity, leading to VSMC hyperplasia. The study suggested that abnormal FAK or Pyk2 activation in vascular cells may cause pathology in vascular diseases. Here we will review several studies of FAK and Pyk2 associated with integrin signaling in vascular diseases including restenosis, atherosclerosis, heart failure, pulmonary arterial hypertension, aneurysm, and thrombosis. Despite the importance of FAK family kinases in vascular diseases, comprehensive reviews are scarce. Therefore, we summarized animal models involving FAK family kinases in vascular diseases.Here we focus on the thermal and variable temperature electrochemical stabilities of two ionic liquids (ILs) having a common tributyloctyl phosphonium cation [P4,4,4,8]+ and two different orthoborate anions bis(mandelato)borate [BMB]- and bis(salicylato)borate [BScB]-. The thermo-gravimetric analysis data suggest that [P4,4,4,8][BScB] is thermally more stable than [P4,4,4,8][BMB] in both nitrogen atmosphere and air, while the impedance spectroscopy reveals that [P4,4,4,8][BScB] has higher ionic conductivity than [P4,4,4,8][BMB] over the whole studied temperature range. In contrast, the electrochemical studies confirm that [P4,4,4,8][BMB] is more stable and exhibits a wider electrochemical stability window (ESW) on a glassy carbon electrode surface as compared to [P4,4,4,8][BScB]. A continuous decrease in the ESWs of both ILs is observed as a function of operation temperature.Functional metal organic framework (MOF) derivatives have attracted tremendous attention as promising catalysts for various reactions. The thermal decomposition strategies have a vital effect on the structures and physicochemical properties of functional MOF derivatives. Nevertheless, what effect does the pyrolysis strategy have on MOF derivatives need further study. In this work, one-step (under dry air) and two-step (first under N2 and then dry air) pyrolysis are chosen to prepare the functional ceria-based MOF derivatives with novel hierarchical pore structure. In comparison with the derivatives prepared by one-step pyrolysis, the two-step pyrolysis composites exhibit better catalytic activity for toluene oxidation due to the higher contents of surface absorbed oxygen species and surface oxygen vacancies. The reusability and durability test demonstrates perfect stability of such functional MOF derivatives. The in-situ UV Raman reveals that two-step strategy is favorable for enhancing the gaseous oxygen activation ability of the functional MOF derivatives. Those findings may instruct the synthesis of functional MOF derivatives via different pyrolysis strategies as well as afford a further understanding of the crucial role of oxygen vacancies.Cardiological diagnostics use maximal and submaximal tests with increasing load. Maximal stress tests are currently considered the gold standard. The Institut für Prävention und Nachsorge, Cologne (IPN) test may be an alternative when maximal patient load is not indicated. The universality of the test is well-documented in sport, but the reliability of this test is unknown. The aim of this study was to assess between-trial and between-day reliability for parameters assessed by the IPN stress test in cardiological patients. In a study of 24 patients aged 39 to 79 years with cardiovascular diseases, the IPN cycle ergometer short test was performed (submaximal performance test). The reliability of heart rate, systolic and diastolic pressure, absolute power at submaximal load, relative performance at submaximal load and target heart rate were assessed. Good (Interclass Correlation Coefficient (ICC) values ranged from 0.832 to 0.894) and excellent (ICC values ranged from 0.904 to 0.969) between-trial reliability was noted. Between-day reliability was good (ICC values from 0.777 to 0.895) and excellent (ICC values from 0.922 to 0.950). The obtained results suggest that the IPN test may be a reliable tool for use in the assessment of cardiological patients, avoiding the implementation of maximal efforts when excessive patient load is not recommended.
