Johnny was fifteen when he was bitten and gained his powers. By sixteen, he was pretty well-established (New York City's first official superhero, but people called him a menace because he hadn't figured out how to prevent casualties and property damage) and had control of his powers.

Peter was nineteen when he went into space with Reed's team (he was Reed's intern, which is why he was asked to go. Johnny wanted to go but realized that the flight would take a long time, and someone had to at least stay to protect the city. He was also nineteen at the time and because of his spiderman thing his grades in high school weren't great, so the team didn't really think he was all that smart) and became irradiated.

During the story, Peter and Johnny are somewhere around 24. Peter has been an established member of the FF for about five years and is exceedingly famous as the hottest and most eligible member of the team. Johnny is well used to his routine as spiderman and is an absolute expert with his powers and abilities and hiding his identity. He often wishes he could tell at least Peter and his sister but he figures they have enough enemies of their own without him adding his own bullshit into the mix.
