Menceritakan tentang duel yang terkenal antara Sherlock Holmes Vs Prof.Moriarty. Prof.Moriarty adalah seorang yang sangat jenius sama seperti Sherlock dan terkait dengan beberapa kasus pembunuhan. Sherlock mengultimatum akan mengalah Prof.Moriarty secara terang-terangan. Pada suatu hari Prof.Moriarty berencana merusak bulan madu sahabat dekat Sherlock, Dr.Watson. Prof. Moriarty menyuruh orang untuk membunuh Dr.Watson ketika perjalanan bulan madu di kereta. Tetapi gagal karena Sherlock diam-diam mengikuti sahabatnya tersbut dan menolongnya.
Pada film ini Sherlock sendiri hanpir meninggal ketika kejadian ia gagal menyusup ke tempat Prof.Moriarty di gedung pembuatan senjata. Ia gagal dan tertangkap bahkan ia sempat di sandra oleh Prof.Moriarty. Sementara Dr.Watson bertarung melawan sniper jitu Kolonel Sebastian Moran. Mereka berhasil kabur dari tempat itu namun sayang, Sherlock terkena ledakan meriam dan sempat berhenti jantungnya. Namun Dr.Watson memberikan suntikan yang merupakan hadiah dari Sherlock sendiri, dan Sherlock berhasil hidup kembali. Sherlock sendiri tahu bahwa tujuan utama Prof.Moriarty adalah memecah belah negara dan saling bermusuhan. Scene Klasik yang terkenal akan terjadi di akhir-akhir film ini, ia jatuh ke jurang bersama Prof.Moriarty ketika sedang bertarung secara sengit.
