Push-To-Fill Autosampler
The push-to-fill autosampler is an automated version of manual injection. A small-volume sample loop can be fitted on an injection valve, while a larger sample loop is mounted on the valve. The push-to-fill autosampler uses a partial-fill mode. The sample needle is a separate part from the connecting tubing, making it a relatively inexpensive part. The sample needle is also replaceable if it becomes blocked. Push-to-fill autosamplers can use the same size syringe for all volume injections.
SmartInject technology improves retention time precision

Thermo Scientific's SmartInject technology eliminates pressure drop associated with injector valve flow cut-off during rotation. The result is improved retention time precision, column lifetime, and reduced interference with analyte detection and solvent front peak quantitation. Figure 6 shows the effect of SmartInject technology. autosampler As a result, retention time precision is increased by up to six-fold.

The 7725 injector's patented SmartInject technology helps improve retention time precision by reducing the injection cycle. This technology enables fast injection cycles and high peak area precision, while the smallest volume of sample required is only 0.7 uL. With a reduced sample volume, SmartInject technology is able to minimize band broadening. The 7725 autosampler can be used for a variety of chromatography applications, including drug discovery, pharmacokinetics, and bioanalytical assays.
Needle-in-loop needle seal

The Autosampler needle-in-looo syringe has a unique design that eliminates the need for a connecting passage to connect the needle and the sample loop. This feature prevents sample from being pushed into the loop when the needle is reloaded. The needle port is a vulnerable spot for sample leakage, with trace amounts of sample deposited in the needle seal region during insertion. To avoid leakage, flush the needle port with water or another liquid to remove any remaining sample.

An autosampler with a needle-in-loop design requires a syringe with a low-pressure needle seal. The needle and connecting tubing are not flushed with mobile phase, meaning the sample will be wasted if it is not injected. As such, the needle-in-loop design is an ideal solution for a variety of analytical applications.
Injection cycle time

In this article, we discuss several strategies that can reduce autosampler injection cycle time and discuss their potential for further development in the years to come. These strategies include increasing the volume of sample, improving column lifetime, and reducing pressure dips that interfere with solvent front peak quantitation and analyte detection. The article includes a case study that illustrates the benefits of these strategies. You can read the full article below.

First, autosampler injection cycle time is affected by the type of sampler. For example, an autosampler that is configured for rapid gradient separation is often faster than a traditional one. The speed of the sampler can be increased by stacking injections, which increases throughput. Moreover, a system with a smaller footprint can increase its throughput. Finally, autosamplers can be configured for open access or multi-sample usage.
Maintenance requirements

The maintenance requirements of autosamplers vary depending on manufacturer. Some require regular replacement of the sampling syringe, while others require a service technician to complete the process. Autosamplers manufactured after 1992 have a liquid detector, which senses the pressure within the pump tubing to know when the liquid has reached the sample pump. To prevent malfunctions, keep the tubing and detector covers clean and securely fastened. Avoid over-tightening the knob on the detector cover as it can cause the cover to crack. Tightening the knob by hand is all that is necessary.

The autosampler consists of two major subunits: a sample injector and a sample compartment. The sample injector delivers a precise aliquot to the column, while the sample compartment stores a large number of sample solutions in vials. Sample trays organize the sample containers. Additional sample capacity is available through optional plate tray chargers. This article explores the maintenance requirements of autosamplers and offers recommendations for selecting and using them.
