Leukotrienes C4 (LTC4), leukotriene D4 (LTD4), and leukotriene E4 (LTE4) are also known as cysteinyl leukotrienes (CysLTs) because they contain cysteinyl groups on C-6. Drugs that block the binding of leuko-trienes to their receptors in lung tissue are under study for the treatment of asthma. A sample solution containing the target analytes is pumped through a stationary phase (LC column) by a mobile phase flowing under high pressure. CysLTs improve the activity of neural pathways associated with the airway and significantly enhance smooth muscle contractile responses to blood vessels. They have a number of physiological roles, including control of smooth muscle contraction, development of inflammation, and inhibition of platelet aggregation. They play a lokaty in smooth muscle contraction in the lungs. They are found in leukocytes (white blood cells) and have three conjugated double bonds; these two facts account for the name. Leukotrienes are a class of highly biologically active inflammatory mediators, which are eicosanoid unsaturated acids with a conjugated triene structure obtained from the lipoxygen metabolism of arachidonic acid.

LTC4 and LTD4 increase airway resistance and vascular permeability, but have little effect on nasal itching and sneezing, and are less capable of inducing nasal overflow than histamine. The metabolic precursor of all prostaglandins is arachidonic acid, a fatty acid that contains 20 carbon atoms and four double bonds. Leukotriene C (Figure 8.35) is a typical member of this group; note the 20 carbon atoms in the carboxylic acid backbone, a feature that relates this compound structurally to arachidonic acid. The production of the prostaglandins from arachidonic acid takes tereny in several steps, which are catalyzed by enzymes. They are classified into six categories according to the nature of their substituents. The leukotriene receptors are divided into two types according to the target cells: BLT receptors activated by LTB4 and CysLTR receptors activated by CysLTs. Leukotrienes are also derived from fatty acids. A group of compounds derived from fatty acids has a wide range of physiological activities; they are called prostaglandins because they were first detected in seminal fluid, which is produced by the prostate gland. Utilized in both plant & animal kingdoms in the biosynthesis of fatty acids (FA), fats & oils signaling molecules derived from FA such as prostaglandins.


The structures of prostaglandins and their laboratory syntheses have been topics of great interest to organic chemists, largely because of the many physi-ological effects of these compounds and their possible usefulness in the phar-maceutical industry. Some of the functions of prostaglandins are control of blood pressure, stimulation of smooth-muscle contraction, and induction of inflammation. LTB4 and LTC4 levels are usually measured in serum specimens, and LTE4 can be measured from urine. The level of LTB4 or LTC4 in the specimen can be determined by double antibody sandwich enzyme immunoassay. Prostaglandins are known to inhibit the aggregation of platelets. A patient seen in the asthma clinic has recorded daily peak flows that are 75% of the baseline.
