1. What are the two research questions during your C. elegans study? (5pts)

2. What are the methods you used to answer the two research questions? (8/ 12 pts)

3. Give the most important reason that C. elegans is used as a model organism in your study? (6pts)
genetically highly conserved to humans.

4. Is UNC-62 a tumor supressor protein or an oncogene? Explain. (8pts)
Tumor supressor. UNC-62 (non-mutated) functions to promote asymmetric cell division. mutation in UNC-62 leads to symmetric cell division in which there is an indefinite increase in cell proliferation, which leads to tumor formation.

5. Is Pbx1/2 an activator or inhibitor of something. A graph is shown. (8pts).
The answer was activator.

6. Researchers need the nematodes to be of synchronised age. Which method can you used to achieve this? (8pts)
Bleaching

7. Explain in DETAIL, a method you could use to determine if a gene has an effect in development of C. elegans. Include controls and why are they used. (12pts)
RNAi or Overexpression

8. Describe the mechanism of RNAi. (12pts)

9. DLG-1 antibody is used to bind to DLG-1 protein and incorporated in the researcher's immunostaining procedure. Why is this a good thing? (12pts)
DLG-1 is a protein in the apical-basal membrane of the seam cells. If antibody's tagged with a fluorophore, it can outline the whole cell, thus rendering it easier to observe development of seam cells.

10. The researchers would like to place/ mix a primary antibody against bla1 and a primary antibody against bla2 in the same tube. Is this possible? If so, how? (12pts)

11. When rif-1 is knocked out, there is an additional number of cells located away from its normal position. Give at least two possible reasons for this. (8pts)
Associated with PAR-proteins.
Rif-1 is similar to rnt-1, it regulates cell proliferation.

12. What is the difference between symmetric and asymmetric cell division? (5pts)
seam cell and non-seam cell fate.
13. Explain in detail ELISA. (5 pts) - The answer was direct ELISA, because it asked for specificity of results.
