These changes in lipid order may be the cause of the inhibition of cholesterol biosynthesis in cells and apoptosis triggered by CBD treatment.
We found that CBD quickly stimulated apoptosis, and dose-dependently in SKN-BE(2) cells of SK-N, with approximately 50 percent of cells dying within 24 hours after being exposed to 40 milligrams CBD. 5A). S4F).
A combination of transcriptomics and metabolomics data also demonstrated that CBD enhances cholesterol biosynthesis in cells through increasing transcription and protein accumulation of the rate-limiting enzyme within the cholesterol biosynthesis pathway, HMGCR. This is in accordance with the findings from phosphoproteomics, which demonstrate that CBD treatment boosts AMP:ATP and activates AMPK Kinase. Additionally, Seahorse extracellular flux assays confirmed that CBD decreases basal oxygen consumption rates in cells, consistent with the AMPK activation.
