Long term effects of lantana (Lantana camera L.) residue incorporation on major soil microbes and on certain soil chemical properties in the rice-wheat cropping system were studied at Palampur (Himachal Pradesh) in an experiment that started in 1988 with four levels of lantana incorporation (0, 10, 20, 30 Mg.ha-1 on fresh weight basis) and three tillage practices (no puddling, puddling, and soil campaction). Among the biotic factors, leaf curl and yellowing disease of watermelon is becoming a major threat for the watermelon production in India.
Genome sequence analysis using Sequence Demarcation Tool (SDT) showed the maximum nucleotide identity of 91.5-93.3% and 80.2-84.5% of six watermelon isolates with Tomato leaf curl Palampur virus (ToLCPalV) for DNA-A and DNA-B components, respectively. Nucleotide BLAST and phylogenetic analyses of the virus isolate showed highest (95%) sequence identity and a close relationships with the isolates of Tomato leaf curl Palampur virus (ToLCPV), suggesting the virus isolate to be the strain of ToLCPV. Complete genome amplification by RCA and sequencing of six isolates was done. RCA products obtained were cloned and sequenced. The amplified product of Gorakhpur isolate was cloned and sequenced, and data was submitted in GenBank. 800 bp product in symptomatic leaf samples of C. pepo collected from all the locations. No amplification resulted in PCR with primers specific to alpha and betasatellites in all the samples. https://pbase.com/topics/drakemail62/agricultural_sector_has_cont -four diseased watermelon samples collected from farmers’ fields during the survey gave positive amplification to begomovirus specific primers in PCR and yielded expected 1.2 kb amplicon, which represent its partial genome. Besides, pH, organic carbon, CEC and available N, P and K were also determined in surface soil samples taken after rice (2008 and 2009) and wheat (2008-09 and 2009-10) using standard methods of analysis.
The soil of the experimental site was silty loam and classified taxonomically as Typic Hapludalf with acidic reaction, medium in organic carbon, high in available N, medium in P and K at the initiation of the experiment. After 12 crop cycles (2001-’02), lantana residue application at 10, 20, and 30 Mg.ha-1 increased soil organic carbon (7, 13, and 19% over 1.29 g C kg-1 under no residue treatment) and pH (5.23 to 5.29 as against 5.12 in the control). Among nitrogen fractions, hydrolysable ammonical-N was found to be the most dominant, which exhibited highest correlation with soil properties, yield and uptake followed by amino acid-N. Incidence of watermelon leaf curl and yellowing disease in ten districts covering different regions of Karnataka State, India varied from 6.8 to 60.7%. Diseased watermelon plants exhibited symptoms typical to begomovirus infection such as leaf crumpling, leaf curling, yellowing, reduction in leaf size and stunted growth in the surveyed fields. Watermelon is important fruit vegetable crop affected by both biotic and abiotic stresses.
