Stem cell therapy is gaining momentum as an effective treatment strategy for degenerative diseases. Adult stem cells isolated from various sources (i.e., cord blood, bone marrow, adipose tissue) are being considered as a realistic option due to their well-documented therapeutic potentials. Our previous studies standardized a method to isolate circulating multipotent cells (CMCs) that are able to sustain long term in vitro culture and differentiate towards mesodermal lineages.
In this work, long-term cultures of CMCs were stimulated to study in vitro neuronal and myogenic differentiation. After induction, cells were analysed at different time points. Morphological studies were performed by scanning electron microscopy and specific neuronal and myogenic marker expression were evaluated using RT-PCR, flow cytometry and western blot. For myogenic plasticity study, CMCs were transplanted into in vivo model of chemically-induced muscle damage.
After neurogenic induction, CMCs showed characteristic dendrite-like morphology and expressed specific neuronal markers both at mRNA and protein level. The calcium flux activity of CMCs under stimulation with potassium chloride and the secretion of noradrenalin confirmed their ability to acquire a functional phenotype. In parallel, the myogenic potential of CMCs was confirmed by their ability to form syncytium-like structures in vitro and express myogenic markers both at early and late phases of differentiation. Interestingly, in a rat model of bupivacaine-induced muscle damage, CMCs integrated within the host tissue taking part in tissue repair.
Overall, collected data demonstrated long-term cultured CMCs retain proliferative and differentiative potentials suggesting to be a good candidate for cell therapy.

Irisin is a unique myokine with striking effects on regulating insulin sensitivity and energy metabolism. This study aimed to investigate the changes in serum irisin in patients with newly diagnosed type 2 diabetes mellitus (T2DM) following sitagliptin treatment.
Thirty-two patients with T2DM were treated with 100mg/day sitagliptin for 16weeks. Twenty age-, sex- and body mass index (BMI)-matched healthy subjects were enrolled as the control group. Irisin and metabolic parameters were measured at baseline and after treatment.
Patients with T2DM had lower irisin levels than the controls (10.03 ± 2.06 vs. 13.06 ± 3.10ng/ml, P < 0.01). Sitagliptin treatment significantly increased serum irisin levels in T2DM patients compared to baseline (11.18 ± 1.91 vs. 10.03 ± 2.06 ng/ml, P < 0.01). link2 Increased irisin levels were associated with decreased fasting blood glucose (FBG) (β = -0.24, P < 0.05) and glycosylated hemoglobin (HbA1c) (β = -0.15, P < 0.05).
Sitagliptin treatment significantly increased serum irisin levels in patients with T2DM, and the increase of the irisin level was associated with decreases of FBG and HbA1c levels. These results suggest that irisin might be involved in the antidiabetic mechanisms of sitagliptin.

Patients with adrenal insufficiency are usually treated with conventional hydrocortisone replacement therapy which fails to mimic the circadian rhythm of cortisol secretion. Dual-release hydrocortisone (DR-HC) resembles the daily normal cortisol profile improving metabolic parameters and quality of life. However, currently little is known about its impact on cognitive function. Aim of this study was to evaluate cognitive function and well-being in DR-HC treated patients compared to healthy controls and conventional HC treatment.
Twenty adults with adrenal insufficiency treated with DR-HC (Plenadren®) underwent 10 neuropsychological tests, evaluating cognitive functions. find more Furthermore, demographic data, quality of life, symptoms of depression, and quality of sleep were evaluated by well-established questionnaires. Patients were compared by diagnosis (PAI/SAI) and dose (≥20 mg). In addition, eighteen DR-HC treated adults were compared to eighteen matched conventionally treated adults.
With respect to diagnosis patients with PAI performed significantly better on intellectual abilities (p = 0.038) and on executive functioning (p = 0.026) and reported a significant longer time to fall asleep (p = 0.026). Regarding DR-HC dosage, there were no significant differences in cognitive functions. Patients on high dose reported a better subjective quality of sleep (p = 0.028) than patients on low dose. In comparison to conventional HC treatment, patients with DR-HC tended to show better results in executive functioning (p = 0.099).
Patients with PAI reached better results in several cognitive functions and had a worse quality of sleep than patients with SAI. Our data suggest a positive impact of DR-HC on quality of sleep. DR-HC may be better for executive functioning.

While there are common normal variations of renal vascular anatomy, such as accessory renal arteries, a main renal artery that originates from the thoracic aorta (thoracic renal artery) is remarkably rare. The majority of reported cases of thoracic renal artery were right side, except two cases reported on 1980 and 2016. The author reports a rare case of left renal artery originating from the thoracic aorta.An increasing number of '-omics' datasets, generated by labs all across the world, are becoming available. link2 They contain a wealth of data that are largely unexplored. Not every scientist, however, will have access to the required resources and expertise to analyze such data from scratch. Fortunately, a growing number of investigators is dedicating their time and effort to the development of user friendly, online applications that allow researchers to use and investigate these datasets. Here, we will illustrate the usefulness of such an approach. We also call out to the mammary gland community to join forces in a coordinated effort to generate and share additional tissue-specific '-omics' datasets and thereby expand the in silico toolbox.Dissolved organic matter (DOM) released from soil (SDOM) and biochar (BDOM) plays an important role in the fate of pollutants for the application of biochar in soil remediation. However, little attention has been paid to the impact of DOM on the adsorption of soil and/or biochar-bisphenol A (BPA) system. Hence, batch sorption experiments combined with quenching titration experiments were processed to reveal these behaviors and complexation mechanisms based on red soil and corn straw biochar. Moreover, multiple spectral technologies and methods were also introduced including ultraviolet-visible, three-dimensional excitation-emission matrix, synchronous fluorescence spectrum, two-dimensional correlation spectroscopy (2D-COS), and parallel factor analysis. Isothermal adsorption results showed that the non-linearity and the adsorption capacity both increased after the addition of biochar, indicating that biochar could improve the surface adsorption capacity of the sorption system. Besides, the sorption capacity of BPA decreased for soil while increased for biochar after removal of the water-extractable DOM. link2 2D-COS analysis showed that the protein-like components preferentially occurred for both SDOM and BDOM under BPA surrounding. In addition, C2 (humic-like), C4 (tyrosine-like), and C5 (tryptophan-like) in the two DOMs were all involved in the quenching reaction with BPA, with C4 and C5 having higher quenching degree than C2, suggesting that protein-like substances had stronger affinity quenching effect with BPA. This study highlighted the significance of fluorescence property for DOM, which can extend the knowledge of the fate of pollutants in the application of biochar.Titanium dioxide (TiO2) nanoparticles (NPs) are employed as an ultraviolet filter in sunscreen products because of their high ultraviolet absorptivity. link3 However, sunscreen sprays may pose health risks due to the toxicity of inhaled TiO2 NPs. Therefore, we estimated the potential human health risk posed by inhaled TiO2 NPs emitted from sunscreen sprays. The physiology-based lung model was employed to predict the lung TiO2 NPs burden caused by long-term exposure. link3 A Hill-based dose-response model described the relationship between lung inflammation and TiO2 NP accumulation. The Weibull threshold model was used to estimate the threshold amount of accumulation inducing 0.5% of the maximum increase in neutrophils. The potential health risk was assessed using a hazard quotient-based probabilistic risk model. All data obtained to date indicate that application of sunscreen sprays poses no significant health risk. link3 However, using data simulations based on the threshold criterion, we discovered that in terms of practical strategies for preventing the risks posed by inhaled TiO2 NPs emitted from spray products, the suggested daily use amount and pressing number are 40 g (95% confidence interval 11-146 g) and 66 (18-245), respectively. In this study, we successfully translated the potential health risk of long-term exposure to NP-containing sunscreen sprays and recommendations for daily application into mechanistic insights.
