LEVEL OF EVIDENCE Level V, expert opinion.We engineered human pancreatic cancer cell (PANC-1)-derived extracellular vesicles (EVs) by conjugating the functional ligand RGD and magnetic nanoparticles (MNPs) onto EV surfaces (rmExo), for pancreatic cancer therapy. Paclitaxel (PTX) loaded into rmExo (rmExo-PTX) was intravenously injected into xenograft mice prepared using PANC-1 cells, which showed a significant reduction in tumor size compared to the free PTX-treated and control groups. The enhanced therapeutic effect was attributed to the modification of the surface of EVs using RGD, which has affinity for αvβ3 that is highly expressed in pancreatic cancer cells. Moreover, autologous EVs seemed to have more benefits in delivering PTX due to an unknown homing property to parent tumor cells, as exemplified by the reduced therapeutic effect of RGD-modified PANC-1 EVs on HT29 xenograft mice and RGD-modified U937 EVs on PANC-1 xenograft mice. The RGD-modified autologous EV vehicles were effective at penetrating and internalizing tumor cells, and eventually regressing the tumors, by mediating spontaneous removal of α-smooth muscle actin and collagen type 1 in the extracellular matrix of xenografts. Our results also identified an important molecule involved in the home-driving properties of PANC-1 EVs, integrin β3, which was expressed both on PANC-1 cells and the EVs derived from them. Additional therapeutic effect by permanent magnet near tumor xenograft was not observed in this study.Cancer vaccines have emerged as powerful and clinically viable therapeutic modalities to reduce tumor burden, eradicate residual cancer cells and prevent relapse. The past years have witnessed rapid advances in various scientific and engineering approaches to next-generation cancer vaccines. This perspective highlights the cutting-edge technologies to elicit robust, durable and cancer-specific immune responses as well as interesting research directions in augmenting the therapeutic efficacies and reducing the systemic side effects of cancer vaccines. The featured technologies include (i) bottom-up high-throughput screening strategies to identify neoantigens as well as optimal delivery systems for tumor antigens and/or adjuvant; (ii) top-down knowledge-based strategies to de novo design effective delivery platforms and to engineer tissue-targeting specificity; and (iii) synergizing cancer vaccines with the clinical immunotherapeutic practices such as CAR-T and anti-PD-1 therapies.This work focuses on obtaining a magnetic resonance imaging (MRI) signal representation that accounts for a longitudinal T1 and transverse T2⋆ relaxations while at the same time integrating directional diffusion in the context of scattered multi-parametric acquisitions, where only a few diffusion gradient directions and b-values are available for each pair of echo and inversion times. The method is based on the three-dimensional simple harmonic oscillator-based reconstruction and estimation (SHORE) representation of the diffusion signal, which enables the estimation of the orientation distribution function and the retrieval of various quantitative indices such as the generalized fractional anisotropy or the return-to-the-origin probability while simultaneously resolving for T1 and T2⋆ relaxation times. Our technique, the Relax-SHORE, has been tested on both in silico and in vivo diffusion-relaxation scattered MR data. The results show that Relax-SHORE is accurate in the context of scattered acquisitions while guaranteeing flexibility in the diffusion signal representation from multi-parametric sequences.Cellulose nanocrystals (CNC) were mixed with either cellulose nanofibril (CNF) or carboxymethylcellulose (CMC) in variable proportions (0/100, 20/80, 40/60, 50/50, 60/40, 80/20 and 100/0) to obtain cast films with acceptable barrier and mechanical properties as replacements for food packaging plastics. Both CNF and CMC improved tensile strength, elongation, UV opacity, air resistance, hydrophobicity (WCA-water contact angle), water vapor transmission rate (WVTR) and oxygen impermeability in pure CNC. WVTR and oxygen permeability were strongly dependent on relative humidity (RH). Interestingly, the greatest effect on WVTR was observed at RH = 90% in films containing CMC in proportions above 60%. CMC- and CNF-containing films had oxygen impermeability up to an RH level of 80% and 60%, respectively. The previous effects were confirmed by food packaging simulation tests, where CMC-containing films proved the best performers. The composite films studied were biodegradable-which constitutes a major environmental related advantage-to an extent proportional to their content in CMC or CNF.The crystal structure of the human telomeric DNA Tel24 G-quadruplex (Tel24 = TAG3(T2AG3)3T) in complex with the novel [AuL] species (with L = 2,4,6-tris(2-pyrimidyl)-1,3,5-triazine - TPymT-α) was solved by a novel joint molecular mechanical (MM)/quantum mechanical (QM) innovative approach. The quantum-refinement crystallographic method (crystallographic refinement enhanced with quantum mechanical calculation) was adapted to treat the [AuL]/G-quadruplex structure, where each gold complex in the binding site was found spread over four equally occupied positions. The four positions were first determined by docking restrained to the crystallographically determined metal ions' coordinates. Then, the quantum refinement method was used to resolve the poorly defined density around the ligands and improve the crystallographic determination, revealing that the binding preferences of this metallodrug toward Tel24 G-quadruplex arise from a combined effect of pyrimidine stacking, metal-guanine interactions and charge-charge neutralizing action of the π-acid triazine. The occurrence of interaction in solution with the Tel24 G-quadruplex DNA was further proved through DNA melting experiments, which showed a slight destabilisation of the quadruplex upon adduct formation.A set of renewable and biodegradable hydrogels based on acid whey and cellulose derivatives blended with poly(lactic acid) (PLA) were designed as eco-friendly biopolymeric material for sustainable agricultural applications. The physico-chemical properties of the hydrogel were evaluated using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and rheological measurements. The effect of the whey/polysaccharide/PLA hydrogel on soil quality improvement (water retention study, biodegradability, loading capacity and release of the fertilizers) and the growth pattern of Raphanus sativus and Phaseolus vulgaris has been also studied. The addition of PLA has been found to improve mechanical properties of the hydrogel. The introduction of 20% wt PLA extended decomposition time of hydrogels by 25% which makes the material more stable in the environment and maintaining the soil humidity for longer. The increasing the amount of PLA led to a rise in hydrogel viscosity brought about better entrapment efficiency of the fertilizers (86-92% for KNO3 and 87-96% for urea, resp.) compared to control (82% for KNO3 and 85% for urea, resp.). The novel hydrogels with swelling ratio of up to 500% showed potential as a sustainable water reservoir for plants improving water retention capacity of the soil by 30%.Active/intelligent packaging films were developed by incorporating purple sweetpotato anthocyanins (PSA) and purple cabbage anthocyanins (PCA) in locust bean gum/polyvinyl alcohol (LP), chitosan/polyvinyl alcohol (CP) and κ-carrageenan/polyvinyl alcohol (KP) film matrices. The joint effect of anthocyanins' source and polysaccharides' nature on the structure and functionality of the films was determined. Results showed PSA and PCA interacted with film matrices through hydrogen bonds and/or electrostatic interactions, resulting in improved film uniformity. PSA and PCA did not remarkably alter the water vapor permeability and tensile strength of the films; however, significantly improved the light barrier ability, antioxidant activity, pH-sensitivity and ammonia-sensitivity of films. For the films containing the same anthocyanins (PSA or PCA), LP-based films had relatively higher light barrier ability and storage stability, while CP-based had relatively weaker color changeable ability. When PSA and PCA were immobilized in the same film matrix, the films containing PCA had higher light barrier ability and antioxidant activity than the films containing PSA. Among the films, LP-PCA film exhibited a good potential to monitor the freshness of shrimp. Results suggested the structure and functionality of the films were influenced by the source of anthocyanins and the nature of polysaccharides.Propionylated potato starch (PPS) with different degrees of substitution (DS) was prepared from native potato starch (NPS) and their potential to encapsulate Lactobacillus rhamnosus GG (LGG) was analyzed. Fourier transform infrared spectroscopy (FTIR) showed a characteristic peak of propionyl groups, which appeared at 1746 cm-1, demonstrating that propionylation occurred. X-ray diffraction (XRD) results revealed that the characteristic diffraction peak intensity of PPS gradually disappeared with the increasing of the DS, which was related to the loss of the ordered crystalline structure of starch granules. Propionylation resulted in the starch to be more thermally stable than its native starch. Furthermore, the propionylated starch had a higher resistance to digestion and hydrophobicity. More importantly, the micro-capsulated LGG derived from propionylated starch could achieve a maximum embedding efficiency of 87.77% at starch DS = 1.54, and also demonstrated a higher resistance to a strong acidic condition and a greater storage stability at 4 °C. This study may highlight a novel approach for probiotic encapsulation using propionylated potato starch as an encapsulant.Misfolding and aggregation of α-synuclein (α-syn) play a key role in the pathogenesis of Parkinson's disease (PD). Herein, the inhibitory effect of ulvan on α-syn fibrillogenesis was studied using thioflavin T fluorescence and atomic force microscope assays. It is shown that ulvan could inhibit α-syn fibrillogenesis in a dose-dependent manner. AU-15330 molecular weight Based on the circular dichroism results, it is found that ulvan delays greatly the conformational transition from its initial random coil to β-sheet rich structure. The protective effect of ulvan against celllular death induced by α-syn aggregates was investigated by MTT colorimetric and cellular staining methods. It is found that ulvan protects greatly PC12 cells from α-syn fibril-induced cytotoxicity. In addition, ulvan disaggregates preformed α-syn fibrils and reduces cytotoxicity in a dose-dependent manner. Thereafter, the inhibitory effects of ulvan against α-syn fibrillogenesis were probed using Caenorhabditis elegans model NL5901 expressing human α-syn. It is found that ulvan extends the lifespan of NL5901 and recovers the lipid deposition by reducing the accumulation of α-syn.
