Craniosynostosis is a condition in which cranial sutures fuse prematurely, causing problems in normal brain and skull growth in infants. To limit the extent of cosmetic and functional problems, swift diagnosis is needed. The goal of this study is to investigate if a deep learning algorithm is capable of correctly classifying the head shape of infants as either healthy controls, or as one of the following three craniosynostosis subtypes; scaphocephaly, trigonocephaly or anterior plagiocephaly. In order to acquire cranial shape data, 3D stereophotographs were made during routine pre-operative appointments of scaphocephaly (n = 76), trigonocephaly (n = 40) and anterior plagiocephaly (n = 27) patients. 3D Stereophotographs of healthy infants (n = 53) were made between the age of 3-6 months. #link# The cranial shape data was sampled and a deep learning network was used to classify the cranial shape data as either healthy control, scaphocephaly patient, trigonocephaly patient or anterior plagiocephaly patient. For find more and testing of the deep learning network, a stratified tenfold cross validation was used. During testing 195 out of 196 3D stereophotographs (99.5%) were correctly classified. This study shows that trained deep learning algorithms, based on 3D stereophotographs, can discriminate between craniosynostosis subtypes and healthy controls with high accuracy. There are indications that pharmacological doses of ascorbate (Asc) used as an adjuvant improve the chemotherapeutic management of cancer. This favorable outcome stems from its cytotoxic effects due to prooxidative mechanisms. Since regulation of intracellular Ca2+ levels contributes to the maintenance of cell viability, we hypothesized that one of the effects of Asc includes disrupting regulation of intracellular Ca2+ homeostasis. Accordingly, we determined if Asc induced intracellular Ca2+ influx through activation of pertussis sensitive Gi/o-coupled GPCR which in turn activated transient receptor potential (TRP) channels in both etoposide-resistant and -sensitive retinoblastoma (WERI-Rb1) tumor cells. Ca2+ imaging, whole-cell patch-clamping, and quantitative real-time PCR (qRT-PCR) were performed in parallel with measurements of RB cell survival using Trypan Blue cell dye exclusion. TRPM7 gene expression levels were similar in both cell lines whereas TRPV1, TRPM2, TRPA1, TRPC5, TRPV4, and TRPM8 gene expresurthermore, Asc may be indicated as a possible supportive agent in anti-cancer therapies. The temperature-dependent mechanical behaviors of open-hole composite plates are essential for composite design and structures. Here, tensile experiments of shallow straight-link-shaped 2.5D woven composites (abbr. 2.5DWC) with/without a center hole are first conducted at different temperatures (20 °C, 180 °C and 240 °C). Failure modes are examined by scanning electron microscope (SEM). Thermal property of QY8911-IV resin is investigated by DMA analysis. It is noted for samples without the center hole that with the increase of temperature, the tensile stress-strain curves exhibit a linear response until that a slight nonlinearity at the end stage. The strength retention rates at 180 °C and 240 °C are totally equal. For the open-hole samples, it is interestingly found that the strength retention rates are higher than that of samples without the hole at 180 °C, resulting from the stress concentration accommodation and fiber-dominated failure mode. Even at 240 °C, there is no necking phenomenon for all the failed samples, but more broom-like damage extent is observed in the cross-section. Due to the primary load-bearing warp yarns and hole-edge stress concentration, obvious pull-out warp yarns emerge near the hole edge. The tibial tuberosity-trochlear groove (TT-TG) distance is a radiographic measurement that is used to quantify malalignment of the patellofemoral joint (PFJ) in cross-sectional imaging. There is an ongoing debate about the impact of the TT-TG-distance on lateral patellar instability and the initiating of cartilage degeneration. In this prospective study, the association of T2* relaxation times and TT-TG distances in professional soccer players was analyzed. 36 knees of 18 professional soccer players (age 21 ± 2.8 years) were evaluated. Participants underwent knee MRI at 3 T. link2 For qualitative image analysis, fat-saturated 2D PD-weighted Fast Spin Echo (FSE) and T1-weighted FSE sequences were used. For quantitative analysis, T2* measurements in 3D data acquisitions were performed. In a qualitative analysis there was no structural cartilage damage and no abnormalities of the patellar and trochlea shape. The highest T2* values (26.7 ± 5.9 ms) were observed in the central compartment of the patella. The mean TT-TG distance was 10 ± 4 mm (range 3-20 mm). There was no significant correlation between TT-TG distance and T2* relaxation times in all three compartments of the retropatellar cartilage. Our study shows that so long as patellar and trochlear morphology is normal, TT-TG distance alone does not affect the tissue structure of the retropatellar cartilage in professional soccer players.Competition between viruses and Wolbachia for host lipids is a proposed mechanism of Wolbachia-mediated virus blocking in insects. link3 Yet, the metabolomic interaction between virus and symbiont within the mosquito has not been clearly defined. We compare the lipid profiles of Aedes aegypti mosquitoes bearing mono- or dual-infections of the Wolbachia wMel strain and dengue virus serotype 3 (DENV3). We found metabolic signatures of infection-induced intracellular events but little evidence to support direct competition between Wolbachia and virus for host lipids. Lipid profiles of dual-infected mosquitoes resemble those of DENV3 mono-infected mosquitoes, suggesting virus-driven modulation dominates over that of Wolbachia. Interestingly, knockdown of key metabolic enzymes suggests cardiolipins are host factors for DENV3 and Wolbachia replication. These findings define the Wolbachia-DENV3 metabolic interaction as indirectly antagonistic, rather than directly competitive, and reveal new research avenues with respect to mosquito × virus interactions at the molecular level.Recent studies have shown how intestinal parasites can modulate gut microbiota. This observation is not surprising since the human intestinal lumen, like any other niche, is a battlefield of microbial competition, and Eukaryotes can affect bacterial populations. Intestinal pathogenic protist has been associated with reshaping the microbial community structure; however, the interactions between the colonic bacterial communities and parasites like Blastocystis spp., Entamoeba coli, and Endolimax nana have been poorly studied. In this work, we studied the distal intestinal bacterial microbiota of 49 children attending 7 public daycare centers in Medellin, Colombia, and compared the bacterial microbiota structure in the presence or absence of the protists Blastocystis spp., E. coli, and E. nana. Parasite colonization was associated with an increase in bacterial richness. Moreover, Blastocystis spp. presented a positive relationship with Prevotella, since this bacterium was selectively enriched in children carrying it. Remarkably, the E. coli colonized children showed a microbial profile that was closer to uninfected controls, although some bacterial taxa displayed to be enriched. This is the case for Akkermansia, which showed to be favored in E. coli colonized individuals, while notably reduced in the Blastocystis spp. parasitized group. An amendment to this paper has been published and can be accessed via a link at the top of the paper. This study deals with an experimental investigation to assess the characteristics of a modified common rail direct injection (CRDI) engine utilizing diesel, Mahua biodiesel, and their blends with synthesized zinc oxide (ZnO) nano additives. The physicochemical properties of diesel, diesel + 30 ppm ZnO nanoparticles (D10030), 20% Mahua biodiesel (MOME20), and Mahua biodiesel (20%) + 30 ppm ZnO nanoparticles (MOME2030) were measured in accordance to the American Society for Testing and Materials standards. The effects of modification of fuel injectors (FI) holes (7-hole FI) and toroidal reentrant combustion chamber (TRCC) piston bowl design on the performance of CRDI using different fuel blends were assessed. For injection timings (IT) and injection opening pressure (IOP) average increase in brake thermal efficiency for fuel blend D10030 and MOME2030 was 9.65% and 16.4%, and 8.83% and 5.06%, respectively. Also, for IT and IOP, the average reductions in brake specific fuel consumption, smoke, carbon monoxide, hydrocarbon and nitrogen oxide emissions for D10030 and MOME2030 were 10.9% and 7.7%, 18.2% and 8.6%, 12.6% and 11.5%, 8.74% and 13.1%, and 5.75% and 7.79%, respectively and 15.5% and 5.06%, 20.33% and 6.20%, 11.12% and 24.8%, 18.32% and 6.29%, and 1.79% and 6.89%, respectively for 7-hole fuel injector and TRCC. The cylinder pressure and heat release rate for D10030 and MOME2030 were enhanced by 6.8% and 17.1%, and 7.35% and 12.28%. The 7-hole fuel injector with the nano fuel blends at an injection timing and pressure of 10° btdc and 900 bar demonstrated the overall improvement of the engine characteristics due to the better air quality for fuel mixing. Similarly, the TRCC cylinder bowl geometry illustrated advanced ignition due to an improved swirl and turbulence. Also, the engine test results demonstrated that 30 ppm of ZnO nanoparticles in Mahua biodiesel (MOME2030) and diesel (D10030) with diethyl ether resulted overall enhancement of CRDI engine characteristics. In this work we apply statistical mechanics tools to infer cardiac pathologies over a sample of M patients whose heart rate variability has been recorded via 24 h Holter device and that are divided in different classes according to their clinical status (providing a repository of labelled data). Considering the set of inter-beat interval sequences [Formula see text], with [Formula see text], we estimate their probability distribution [Formula see text] exploiting the maximum entropy principle. By setting constraints on the first and on the second moment we obtain an effective pairwise [Formula see text] model, whose parameters are shown to depend on the clinical status of the patient. In order to check this framework, we generate synthetic data from our model and we show that their distribution is in excellent agreement with the one obtained from experimental data. Further, our model can be related to a one-dimensional spin-glass with quenched long-range couplings decaying with the spin-spin distance as a power-law. This allows us to speculate that the 1/f noise typical of heart-rate variability may stem from the interplay between the parasympathetic and orthosympathetic systems.Regulating the amount of proteins in living cells is a powerful approach for understanding the functions of the proteins. Immunomodulatory drugs (IMiDs) induce the degradation of neosubstrates by interacting with celebron (CRBN) in the cullin E3 ubiquitin ligase complex (CRL4CRBN). Here, we developed the IMiD-dependent Sal-like protein 4 (SALL4) degron (S4D) system for chemical protein knockdown. In transient assays, an N- or C-terminal S4D tag induced the degradation of proteins localized to various subcellular compartments, including the plasma membrane. The activity of luciferase-S4D was reduced by 90% within 3 h of IMiD treatment. IMiD treatment reduced the expression of endogenous S4D-fused RelA and IκBα in knock-in (KI) experiments. Interestingly, the IκBα knockdown suggested that there may be another, unknown mechanism for RelA translocation to the nucleus. Furthermore, 5-hydroxythalidomide as a thalidomide metabolite specifically degradated S4D-tagged protein. These results indicate that the S4D system is a useful tool for cellular biology.
